新生期七氟烷麻醉对C57BL/6雄性小鼠社会交往行为的影响及机制探讨

Effect of Neonatal Anesthesia with Sevoflurane on Social Interaction Behavior in C57BL/6 Male Mice and Its Potential Mechanism

目的探索新生期七氟烷麻醉对C57BL/6雄性小鼠社会交往行为的影响,并探讨其潜在机制。方法在SPF级实验动物房培育子代小鼠,将出生后第6天的雄性小鼠随机分配至两组进行处理:七氟烷组小鼠吸入3.0%七氟烷+60%氧气(10只),对照组小鼠单纯吸入60%氧气(10只),时间均为2 h。在幼年期(出生后30天)和成年早期(出生后60天),测试小鼠的社会交往行为。繁殖另一批雄性子代小鼠,将其随机分配至对照组、氧气组和七氟烷组(每组4只),获取麻醉处理后小鼠大脑皮层和海马组织进行免疫印迹分析。结果在幼年期,七氟烷组雄性小鼠表现较强的社会交往倾向和较弱的社会新奇偏好。在成年早期,七氟烷组雄性小鼠表现较强的社会交往倾向和正常的社会新奇偏好。与对照组比较,七氟烷组新生期小鼠海马PSD95蛋白表达水平显著性降低。结论新生期七氟烷麻醉可降低雄性小鼠海马PSD95蛋白表达水平,但是不会直接损害雄性小鼠幼年期和成年早期的社会交往行为。

Objective To investigate whether sevoflurane anesthesia in neonatal period could impair social interaction behaviors in C57BL/6 male mice and its potential mechanism.Methods The offspring mice were produced in the SPF animal room,and the six-day-old male mice were randomly assigned to two groups,and then they were anesthetized with 3.0%sevoflurane pluse 60%oxygen for 2 h(Sevoflurane,n=10)or treated with just 60%oxygen for 2 h(Control,n=10).These mice were tested for social interaction behaviors at the juvenile(postnatal day 30)and early-adult(postnatal day 60)age.Another battery of neonatal male mice was produced and randomly assigned into three groups of the Control,Oxygen and Sevoflurane(n=4 in each),and their brain tissues including cortex and hippocampus were harvested for immunoblotting analysis.Results The male mice undergoing sevoflurane anesthesia showed strong sociability and weak preference for social novelty at the juvenile age.Moreover,these male mice developed robust sociability,as well as normal preference for social novelty at the early-adult age.The neonatal mice undergoing sevoflurane anesthesia showed lower levels of PSD95 protein in the mouse hippocampus,as compared with the control.Conclusion Neonatal anesthesia with sevoflurane could significantly decrease the levels of hippocampal PSD95 protein.However neonatal exposure to anesthesia could not disturb social interaction behavior in male mice at the juvenile or early-adult age.
