铜绿假单胞菌PvdD蛋白主要特性及抗原表位的生物信息学分析

摘要 为了分析铜绿假单胞菌中假单胞菌素合成酶D(PaPvdD)蛋白的主要特性与抗原表位,进而为研制基于合成酶D(PvdD)的铜绿假单胞菌疫苗提供依据,本研究首先从NCBI蛋白质数据库中下载PaPvdD蛋白的氨基酸序列,然后采用ProtParam、SignalP 4.1、PSORTb 3.0、MotifScan、SYFPEITHI等在线分析软件,结合DNAMAN、DNAStar等生物信息学软件分析、预测PaPvdD蛋白的理化性质、信号肽序列、亚细胞定位、翻译后修饰位点以及B、T细胞表位。结果表明,PaPvdD蛋白由2448氨基酸(aa)组成,是一种亲水性的不稳定蛋白,不含有信号肽序列和跨膜结构域,可能定位于细胞质。PaPvdD蛋白含有3个B细胞表位(分别位于687~723 aa、1748~1784 aa、2194~2210 aa)和5个T细胞表位(分别位于97~105 aa、152~160 aa、697~705 aa、787~795 aa、1005~1013 aa)。本研究通过生物信息学方法筛选出了PaPvdD蛋白的3个B细胞表位和5个T细胞表位,这为研制基于PvdD的铜绿假单胞菌疫苗提供了理论基础。 In order to analyze the main molecular characteristics and epitopes of pyoverdine synthetase D from Pseudomonas aeruginosa(PaPvdD),which will lay foundation for the production of Pa vaccines based on PvdD.In this study,the amino acid sequence of PaPvdD protein was downloaded from protein database on NCBI.Then the bioinformatics tools,such as ProtParam,SignalP 4.1,PSORTb 3.0,MotifScan,SYFPEITHI,were used to analyze physiological.Also the biochemical characteristics,signal peptide sequence,sub-cellular localization and post-translational modification sites.T-and B-cell epitopes were predicted by SYFPEITHI,DNAStar and DNAMAN.The results showed that PaPvdD is a hydrophilic unstable protein,which consisted of 2448 amino acids and locates in cytoplasm.No signal peptide sequence and transmembrane domains were found in the amino acid sequence of PaPvdD.PaPvdD protein is predicted to contain three B-cell epitopes(located at amino acids 687~723,1748~1784,and 2194~2210)and five T-cell epitopes(located at amino acids 97~105,152~160,697~705,787~795,1005~1013).In this study,three B-cell epitopes and five T-cell epitopes of PaPvdD protein were screened by bioinformatics,which lay foundations for the production of Pa vaccines based on PvdD.
