摘要
目的优选砂烫诃子炮制工艺及砂烫诃子中鞣花酸提取工艺。方法采用高效液相色谱法,以炮制温度、炮制时间、翻炒频率为考察因素,以鞣花酸含量为考察指标,采用L_(9)(3^(4))正交试验优化诃子砂烫炮制工艺;以粒径、液料比、提取时间、提取次数为考察因素,以出膏率、鞣花酸提取量、膏中纯度为考察指标,采用L_(16)(4^(5))正交试验设计优选砂烫诃子中鞣花酸提取工艺,采用层次分析法、指标重复性相关法、混合加权法确定指标权重系数,并对各种指标进行综合评分。结果诃子最佳炮制工艺为260℃下砂烫6 min,翻埋频率40次/min。经3次验证,鞣花酸的平均含量为32.5 mg·g^(-1),RSD为1.47%(n=3);砂烫诃子中鞣花酸最佳提取工艺为粒径120目,液料比10∶1,提取时间24 h,提取次数4次。经3次验证,平均综合评分为99.37,RSD为0.19%(n=3)。结论优选所得诃子砂烫炮制工艺及砂烫诃子中鞣花酸提取工艺稳定可靠,可用于诃子的炮制及鞣花酸的提取。
Objective The processing technology and extraction technology of ellagic acid from Terminalia chebula by sand-scorch were optimized.Methods Using high performance liquid chromatography,processing temperature,processing time and digging in frequency as factors and ellagic acid content as index,the processing technology of T.chebula was optimized by L_(9)(3^(4))orthogonal test.With particle size,liquid ratio,extraction time,extraction times to examine factors,in order to extract rate,ellagic acid extraction quantity,purity as examining index in the cream,adopting L_(16)(4^(5))orthogonal experiment design optimization in the hot sand myrobalan ellagic acid extraction technology,using analytic hierarchy process(AHP),index of repeatability correlation method,mixed weighting method to determine index weight coefficients,and the comprehensive score of various indicators.Results The optimal processing technology of T.chebula was sand-scorching at 260℃for 6 min and digging in frequency was 40 times·min^(-1).After three tests,the average content of ellagic acid was 32.5 mg·g^(-1)with RSD of 1.47%(n=3).The optimal extraction conditions of ellagic acid from T.chebula were as follows:particle size 120 mesh,ratio of liquid to material 10∶1,extraction time 24 h,extraction times 4 times.The mean comprehensive score was 99.37 and RSD was 0.19%(n=3).Conclusion The optimized processing technology of T.chebula and the extraction technology of ellagic acid from T.chebula are stable and reliable,and can be used for processing T.chebula and extracting ellagic acid.
