等温扩增荧光技术快速检测沙门氏菌的研究

摘要 本文研究并建立等温扩增荧光技术快速检测沙门氏菌的方法。通过对沙门氏菌的SSAQ基因序列保守区设计引物,采用最佳引物组搭配其他配套试剂建立一种针对沙门氏菌的等温扩增荧光技术快速检测方法;采用直扩法提取DNA,通过等温扩增荧光技术与常规PCR法对不同浓度沙门氏菌进行扩增检测,检测最低检测限,对其灵敏度进行评价。结果表明,所建立的针对沙门氏菌等温扩增荧光检测的方法与常规PCR法相比,等温扩增荧光检测的方法灵敏度更高,且根据干扰实验结果可知该方法对沙门氏菌具有良好的特异性,所建立的针对沙门氏菌的等温扩增荧光技术检测方法具有操作快捷、简便、反应时间较短、良好的特异性和较高的灵敏度的特点,能够用于沙门氏菌的快速检测。 A method for rapid detection of Salmonella by isothermal amplification fluorescence was developed.By designing primers for the conserved region of SSAQ gene sequence of Salmonella,an isothermal amplification fluorescence method for rapid detection of Salmonella was established by combining the best primers with other supporting reagents.The direct expansion method was used to extract DNA,and the isothermal amplification fluorescence technique and conventional PCR method were used to detect Salmonella with different concentrations.The minimum detection limit was detected,and the sensitivity was evaluated.The results showed that the established method for isothermal fluorescence amplification detection of Salmonella is more sensitive than the conventional PCR method,and it has good specificity for Salmonella based on interfering experimental results.The characteristics of this method are fast,simple,short reaction time,good specificity,and high sensitivity.It can be used for rapid detection of Salmonella.
