心衰康胶囊对糖尿病大鼠肾脏保护作用及其机制初步研究

摘要 目的验证心衰康胶囊对糖尿病大鼠肾脏的保护作用并初步研究其可能机制。方法将28只无特定病原体级健康雄性SD大鼠随机分为正常对照组7只和模型组21只。模型组大鼠采用高脂饲料及尾静脉注射链脲佐菌素致糖尿病大鼠模型,模型制备成功后,随机分为模型组、心衰康胶囊组、阳性药物缬沙坦组,每组7只。心衰康胶囊按照1 g·kg^(-1)·d^(-1)的剂量、阳性药物缬沙坦组按照30 mg·kg^(-1)·d^(-1)剂量均灌胃10周。对照组及模型组灌胃0.9%氯化钠注射液,定期测定各组大鼠血糖、体质量,采用苏木素-伊红染色和马松染色观察肾组织病理变化;免疫组织化学法测定肾组织中转化生长因子β1(TGF-β1)蛋白表达水平。结果与正常组比较,糖尿病组大鼠体质量增加缓慢,肾脏组织中TGF-β1表达明显上调(P<0.01),肾组织可见明显胶原组织沉积、纤维化改变,肾结构破坏明显;与模型组比较,心衰康组糖尿病大鼠体质量增长较快(P<0.05),肾组织中TGF-β1表达明显减少(P<0.01),胶原组织沉积减少、纤维化减轻,肾结构破坏不明显。结论心衰康胶囊可能通过抑制糖尿病大鼠肾脏TGF-β1表达,从而减缓糖尿病大鼠肾间质改变,保护改善肾功能。 Objective To verify the protective effect of Xinshuaikang capsules on the kidneys of diabetic rats and to preliminarily investigate its possible mechanism.Methods Twenty-eight healthy SPF male SD rats were randomly divided into the normal control group(n=7)and model group(n=21).The rats in the model group were treated with high-fat feed and tail vein injection of streptozotocin to cause diabetes,and after the model was successfully prepared,they were randomly divided into the model group,Xinshuaikang capsules group,and positive drug valsartan group,with 7 mice per group.Xinshuaikang capsules according to the dose of 1g·kg^(-1)·d^(-1),the positive drug valsartan group according to 30 mg·kg^(-1)·d^(-1) dose were gastric filling 10 weeks.The control group and the model group were filled with gastric normal saline,the blood glucose and body weight of rats in each group were measured regularly,and the pathological changes of renal tissue were observed by hematin-mixin(HE)staining and Masson staining.The expression level of transforming growth factor(TGF)-β1 protein in renal tissue was detected with immunohistochemistry.Results Compared with the normal group,the weight gain of rats in the diabetic group was slow,the expression of TGF-β1 in renal tissue was significantly upregulated(P<0.01),and obvious collagen tissue deposition and fibrosis changes were detected in kidney tissue,and the renal structure was obviously destroyed.Compared with the model group,the diabetic rats in the Xinshuaikang capsules group had faster weight gain(P<0.05),the expression of TGF-β1 in renal tissue was significantly reduced(P<0.01),collagen tissue deposition was reduced,fibrosis was reduced,and renal structure destruction was not obvious.Conclusion Xinshuaikang capsules may inhibit the expression of TGF-β1 in diabetic rats,thereby slowing down the renal interstitial changes in diabetic rats and protecting and improving renal function.
