摘要
目的:借助生物信息学技术分析GEO数据库中新型冠状病毒感染心肌细胞的基因表达数据,尝试探究其分子机制,并预测治疗的潜在中药。方法:在GEO数据库中获得新型冠状病毒感染心肌细胞的基因表达谱数据集GSE185602,利用Excel软件进行分析,筛选差异基因,并进行蛋白质-蛋白质相互作用网络分析,筛选核心基因。对差异基因进行富集分析,获得相关信号通路。结合核心基因和信号通路分析疾病的分子机制。将核心基因与coremine medica数据库映射,获得治疗的潜在中药。结果:研究获得差异基因共232个,其中上调基因128个、下调基因92个,富集分析获得20条信号通路,映射到潜在治疗中药65味。结论:本研究发现新型冠状病毒感染患者继发心肌损伤可能参与补体和凝血级联、细胞色素P450(Cytochrome P450,CYP)代谢异生素和磷脂酰肌醇3激酶(Phosphatidylinositol 3-kinase,PI3K)-丝氨酸/苏氨酸蛋白激酶(RAC-alpha Serine/Threonine-protein Kinase,Akt)信号通路发生,多种中药可具有潜在治疗作用,注意随证选用。
Objective:To analyze the gene expression data of myocardial cells infected with COVID-19 in GEO database by means of bioinformatics technology,try to explore its molecular mechanism,and predict the potential TCM medicine for treatment.Methods:The gene expression profile data set GSE185602 of myocardial cells infected with COVID-19 was obtained from GEO database,and the Excel software was used to analyze and screen the differential genes,and the protein interaction network was analyzed to screen the core genes.The differential genes were enriched and analyzed to obtain relevant signal pathways.The molecular mechanism of disease was analyzed by combining core genes and signal pathways.The core gene was mapped with the coremine medica database to obtain the potential TCM medicine for treatment.Results:A total of 232 differential genes were obtained,including 128 up-regulated genes and 92 down-regulated genes.Twenty signal pathways were obtained by enrichment analysis.There were 65 potential therapeutic TCM medicines.Conclusion:This study found that the secondary myocardial injury in patients with COVID-19 may be involved in the complement and coagulation cascade,cytochrome P450 metabolism allobiotin and PI3K-Akt signal pathway,and a variety of TCM medicines can have potential therapeutic effects,and attention should be paid to the selection of the medicine according to the syndrome.
