摘要
温州病毒(WENV)是新发现、具有潜在致病性的沙粒病毒,监测其流行状况,有利于维护公共卫生安全。对病毒核衣壳蛋白(N)进行了生物信息学分析,并在体外诱导表达及鉴定。结果显示,N蛋白是一种亲水性稳定蛋白,高级结构以无规则卷曲为主,存在磷酸化、糖基化位点及抗原表位;诱导的N蛋白约64 kDa,主要以包涵体的形式存在,纯化后的浓度可达0.62 mg/mL,成功构建了重组原核表达质粒p ET-30a(+)-Np,为WENV检测方法的建立奠定基础。
Wenzhou virus(WENV)is a newly discovered and potentially pathogenic sand virus.Monitoring its epidemic status is conducive to the maintenance of public health and safety.In this study,viral nucleocapsid protein(N)was analyzed by bioinformatics,and induced expression and identification in vitro.The results showed that N protein is a hydrophilic stable protein with a higher structure mainly characterized by irregular curls and the presence of phosphorylation,glycosylation sites,and antigenic epitopes.The induced N protein is about 64 kDa,mainly in the form of inclusion bodies,and the purified concentration can reach 0.62 mg/mL.Therefore,the recombinant prokaryotic expression plasmid pET-30a(+)-Np was successfully constructed in this study,which laid the foundation for the establishment of WENV detection method.
