基于Wnt信号通路对益气通痹胶囊治疗颈动脉粥样硬化作用机制研究

摘要 目的研究并探讨益气通痹胶囊对颈动脉粥样硬化大鼠Wnt信号通路及动脉硬化的影响及作用机制。方法采用随机数字表法将50只大鼠分为高脂模型组、对照组、益气通痹胶囊高浓度组、益气通痹胶囊中浓度组、益气通痹胶囊低浓度组,分别以临床剂量2倍、临床等剂量、临床剂量0.5倍喂食益气通痹胶囊。采用化学酶检测法检测大鼠血脂中TC、TG、LDL-C和HDL-C水平;RT-PCR法检测大鼠颈主动脉组织β-catenin、GSK-3β、VEGF基因表达水平。结果高脂模型组大鼠与对照组相比,血清中TC、TG、LDL-C表达水平显著增高,HDL-C水平显著降低(P<0.05),颈主动脉组织中β-catenin、GSK-3β、VEGF基因表达水平升高;各治疗组大鼠与模型组相比,血清中TC、TG、LDL-C水平降低,HDL-C水平显著升高(P<0.05),颈主动脉组织中β-catenin、GSK-3β、VEGF基因表达水平降低。结论益气通痹胶囊可通过抑制Wnt信号通路中β-catenin、GSK-3β、VEGF基因的表达降低大鼠颈动脉粥样硬化程度,抑制动脉粥样硬化大鼠血管新生。 Objective To study the effect of Yiqi Tongbi capsule on Wnt signaling pathway and arteriosclerosis in rats with carotid atherosclerosis,and to explore the mechanism of Yiqi Tongbi capsule in preventing and treating carotid atherosclerosis.Methods Fifty rats were randomly divided into high-fat group,control group,high-concentration group,medium-concentration group,and low-concentration group,and high-fat diet was used as a mutagen to establish a rat carotid atherosclerosis model.After 16 weeks,blood was collected and sacrificed.Chemoenzymatic method was used to detect the levels of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)in blood lipids of rats,gene expression levels ofβ-catenin,GSK-3βand VEGF in mouse carotid aorta.Results Compared with the control group,the levels of TC,TG,LDL-C and the gene expression levels ofβ-catenin,GSK-3βand VEGF in the carotid aorta of the model group were increased,and the level of HDL-C was significantly decreased(P<0.05).Compared with the model group,the levels of TC,TG,LDL-C and the gene expression levels ofβ-catenin,GSK-3βand VEGF in carotid aorta tissue were decreased in each treatment group,and the level of HDL-C was significantly increased(P<0.05).Conclusion Yiqi Tongbi capsule can reduce the degree of carotid atherosclerosis in rats by inhibiting the expression ofβ-catenin,GSK-3βand VEGF genes in the Wnt signaling pathway,and inhibit angiogenesis in rats with atherosclerosis.
