木犀草苷对运动性疲劳大鼠的心肌氧化应激及抗氧化基因表达的影响 被引量:2

Effects of Cynaroside on Myocardial Oxidative Stress and Expression of Antioxidant Genes in Exercise-Induced Fatigue Rats

摘要 木犀草苷(cynaroside,CYN)是一种存在与多种植物中的黄酮类化合物,具有显著的抗氧化活性。本研究旨在揭示木犀草苷对运动性疲劳诱导的大鼠心肌氧化应激的影响。本研究通过递增负荷游泳建立了运动性疲劳大鼠模型,游泳当天对大鼠分别灌胃2 mL 5 mg/kg、10 mg/kg和20 mg/kg的木犀草苷,连续9周。末次游泳24 h后,检测各组大鼠的心功能参数、血清心肌损伤指标和心肌组织氧化应激指标。通过HE染色观察心肌组织学组织形态。通过RT-qPCR检测心肌组织中Nrf2、Keap1、HO-1和NQO-1的m RNA水平。通过Western blot检测心肌组织中Nrf2的核转位。结果表明,木犀草苷降低了运动性疲劳大鼠的LVPWd、LVPWs和LVIDs,升高了EF和FS(P<0.05)。木犀草苷降低了运动性疲劳大鼠血清LDH、CK和cTnI水平,减轻了心肌组织学病变(P<0.05)。木犀草苷升高了运动性疲劳大鼠心肌组织SOD、GSH-Px和CAT活性,降低了MDA含量(P<0.05)。木犀草苷升高了运动性疲劳大鼠心肌组织Nrf2、HO-1和NQO-1 m RNA相对表达量,降低了Keap1 m RNA相对表达量(P<0.05)。木犀草苷促进了运动性疲劳大鼠心肌组织Nrf2的核转位(P<0.05)。本研究表明木犀草苷可能通过激活Nrf2途径减轻了运动性疲劳大鼠的心肌损伤和氧化应激反应。木犀草苷在预防运动性疲劳或运动性心肌损伤方面具有潜在的应用价值。 Cynaroside(CYN)is a kind of flavonoids found in many plants,which has significant antioxidant activity.This study aimed to reveal the effect of cynaroside on exercise-induced fatigue(EIF)-induced myocardial oxidative stress in rats.In this study,the exercise-induced fatigue rat model was established by swimming with in creasing load.On the day of swimming,2 mL of 5 mg/kg,10 mg/kg and 20 mg/kg cynaroside were administered to the rats for 9 consecutive weeks.24 hours after the last swim,the cardiac function parameters,serum myocardial injury indicators and myocardial tissue oxidative stress indicators of rats in each group were detected.Myocardial histology was observed by HE staining.The mRNA levels of Nrf2,Keap1,HO-1 and NQO-1 in myocardial tissue were detected by RT-qPCR.The nuclear translocation of Nrf2 in myocardial tissue was detected by Western blot.The results showed that cynaroside reduced LVPWd,LVPWs and LVIDs of exercise-induced fatigue rats,and increased EF and FS(P0.05).Cynaroside reduced the levels of serum LDH,CK and cTnI in exercise-induced fatigue rats,and alleviated myocardial histological lesions(P0.05).Cynaroside increased the activities of SOD,GSH-Px and CAT in myocardial tissue of exercise-induced fatigue rats,and decreased the content of MDA(P0.05).Cynaroside incre-ased the relative expression of Nrf2,HO-1 and NQO-1 mRNA in myocardial tissue of exercise-induced fatigue rats,and decreased the relative expression of Keap1 m RNA(P0.05).Cynaroside promoted the nuclear translocation of Nrf2 in myocardial tissue of exercise-induced fatigue rats(P0.05).This study shows that cynaroside may reduce myocardial injury and oxidative stress in exercise-induced fatigue rats by activating the Nrf2 pathway.Cynaroside has potential application value in preventing exercise-induced fatigue or exercise-induced myocardial injury.
