摘要
建立了使用离子交换色谱法检测调制乳粉和固体饮料中异麦芽糖、异麦芽三糖、潘糖的方法。通过β-半乳糖苷酶酶解乳糖,调节p H值沉淀蛋白并提取,采用CarboPacTM PA10阴离子交换柱(4.6 mm×250 mm),以醋酸钠和氢氧化钠为淋洗液进行二元梯度洗脱,脉冲安培法进行检测,测得异麦芽糖、异麦芽三糖、潘糖的含量。该方法重复性和线性关系良好,异麦芽糖、异麦芽三糖、潘糖的回收率为88.5%~101.9%、90.1%~105.9%、88.6%~103.7%,相对标准偏差(RSD)为6.19%、4.89%、5.43%,检出限为0.0040、0.0065、0.0080 g/100 g,该方法可准确测定调制乳粉和固体饮料中异麦芽糖、异麦芽三糖、潘糖的含量。
Methods for determination of isomaltose,isomaltotriose,panose in formulated milk powder and solid beverages by high performance ion exchange chromatography was established and compared.Proteins in the sample was precipitated by adjusting pH to 4.5,and lactoses was enzymolisised byβ-galactosidase,separated on CarboPacTM PA 10 column with a gradient elution of NaOH-NaOAc as the mobile phase and coupledwithpulsedamperometric detection.The content of the isomaltooligosaccharides was calculated by the concentration of isomaltose,isomaltotriose,and panose.The recovery were 88.5%-101.9%、90.1%-105.9%、88.6%-103.7%,the relative standard deviations(RSD)were 6.19%、4.89%、5.43%,the minimum detectable values were 0.0040、0.0065、0.0080 g/100g.This method was accurate and stable to determine the concentration of isomaltose,isomaltotriose,panose in formulated milk powder and solid beverages.
