子宫内膜癌中LncRNA FAM83H-AS1的表达以及与患者临床病理特征和预后的关系

Expression of LncRNA FAM 83H-AS1 in endometrial carcinoma and its relationship with clinicopathological characteristics and prognosis

目的:检测人子宫内膜癌组织及癌旁组织中LncRNA FAM83H-AS1的表达水平,并通过细胞学实验验证LncRNA FAM83H-AS1对子宫内膜癌细胞增殖、侵袭和转移的影响。方法:通过细胞增值实验、细胞克隆形成实验、EdU染色、流式细胞术、Transwell迁移以及侵袭实验等方法分析在过表达或者敲低LncRNA FAM83H-AS1的子宫内膜癌细胞的生长增殖能力与凋亡水平的变化。结果:(1)LncRNA FAM83H-AS1在子宫内膜癌组织中的表达水平明显增高;(2)敲低LncRNA FAM83H-AS1削弱子宫内膜癌细胞的增殖活性;(3)敲低LncRNA FAM83H-AS1促进子宫内膜癌细胞的凋亡;(4)敲低LncRNA FAM83H-AS1抑制子宫内膜癌细胞的迁移与侵袭。结论:LncRNA FAM83H-AS1阳性可能与子宫内膜癌的发生发展有关,并可能与促进癌细胞的转移有关。

Objective:To detect the expression level of IneRNA FAM83H-ASI in human endometrial cancer tissues and adjacent tissues,and verifty the effect of IncRNA FAM83H-ASI on the proliferation,invasion and metastasis of endometrial cancer cells through cytological experiments.Methods:The growth and proliferation ability and apoptosis level of endometrial cancer cells overexpressing or knockdown L.ncRNA FAM83H-ASI were analyzed by cell proliferation test,cell clone formation test,EdU staining,flow cytometry,Transwell migration and invasion test.Results:(1)The expression level of LncRNA FAM83H-AS1 in endometrial carcinoma was significantly increased;(2)Knocking down LncRNA FAM83H-ASI weakened the proliferation activity of endometrial carcinoma cells;(3)Knockdown L.ncRNA FAM83H-AS1 promotes apoptosis of endometrial carcinoma cells;(4)Knockdown of LncRNA FAM83H-ASI inhibits the migration and invasion of endometrial cancer cells.Conclusion:The positive expression of L.ncRNA FAM83H-ASI may be related to the occurrence and development of endometrial carcinoma,and may be related to the promotion of cancer cell metastasis.
