锦灯笼配方颗粒UPLC指纹图谱研究 被引量:2

摘要 目的建立锦灯笼配方颗粒的超高效液相色谱(UPLC)指纹图谱,为锦灯笼配方颗粒的质量控制和评价提供参考依据。方法采用CORTECS UPLC T3(2.1 mm×100 mm,1.6μm)色谱柱,乙腈(A)和0.2%磷酸水溶液(B)为流动相,进行梯度洗脱,检测波长220 nm,流速0.30 mL·min^(-1),柱温25℃,建立锦灯笼配方颗粒UPLC指纹图谱。结合相似度、SPSS系统聚类分析和主成分因子分析,评价不同产地药材制备的锦灯笼配方颗粒的质量差异。结果建立了锦灯笼配方颗粒指纹图谱,标定了11个共有特征峰,并指认了其中5个峰,峰1为木犀草苷、峰3为木犀草素、峰4为酸浆苦味素A、峰6为酸浆苦味素L、峰10为4,7-二脱氢新酸浆苦素B。相似度评价结果0.984~1.000。系统聚类分析结果显示15批不同产地药材样品制得的配方颗粒聚为4类,提取到两大主成分,累积方差贡献率高达88.022%,其中木犀草苷和酸浆苦味素A是对锦灯笼配方颗粒质量影响较大的成分。结论建立的锦灯笼配方颗粒指纹图谱方法简便、耐用性好,能够全面、真实地反映锦灯笼配方颗粒的质量差异,为锦灯笼配方颗粒质量控制及整体性评价提供参考。 Objective The ultra-high performance liquid fingerprint(UPLC)fingerprint method was established to provide reference for the quality control and evaluation of Physalis Calyxseuf Ructus formula Granules.Methods Gradient elution was performed on a CORTECS UPLC T3(2.1 mm×100 mm,1.6μm)column with acetonitrile(A)and 0.2%phosphoric acid(B)as mobile phase.The detection wavelength was 220 nm,the flow rate was 0.30 mL per minute,and the column temperature was 25℃.The UPLC of Physalis Calyxseuf Ructus was established.Similarity,SPSS system cluster analysis and principal component factor analysis were used to compare and analyze the quality differences of Physalis Calyxseuf Ructus formula Granules prepared from different places of origin.Results Eleven common characteristic peaks were selected and 5 peaks were identified.Peak 1 was luteolin-7-O-glucoside,Peak 3 was luteolin,Peak 4 was physalin A,Peak 6 was physalin L,Peak 10 was 4,7-didehydroneophysalin B.The similarity was between 0.984 and 1.000.The systematic cluster analysis showed that 15 batches of medicinal materials samples from different places were clustered into 4 categories,two main components were extracted,and the cumulative variance contribution rate was as high as 88.022%.Luteolin-7-Oglucoside and physalin A were the main components which had great influence on the Physalis Calyxseuf Ructus formula granules quality.Conclusion The established fingerprinting method of Physalis Calyxseuf Ructus formula granules is simple and good repeatability,which can comprehensively and truly reflect the quality difference of Physalis Calyxseuf Ructus formula granules,providing a reference for quality control and overall evaluation of Physalis Calyxseuf Ructus formula granules.
