HPLC-MS法测定阿胶益寿口服液中驴源多肽的含量

Content Determination of Peptide in Ejiao Yishou Oral Liquid by HPLC-MS

为了建立超滤离心纯化处理-液相色谱-质谱联用法同时测定阿胶益寿口服液中驴源多肽A1、驴源多肽A2含量的方法,采用胰蛋白酶水解、3kD超滤离心管纯化、液质联用法,利用质谱仪(采用Waters Symmetry C18(150 mm×2.1 mm,5μm)色谱柱,流动相乙腈、0.1%甲酸梯度洗脱)同时检测了2种特征肽的含量。研究发现,驴源多肽A1、驴源多肽A22种成分的线性关系良好(r≥0.9965),加样回收率平均值分别为101.13%和88.54%,RSD分别为4.19%和6.63%。该方法可实现含阿胶中成药的快速、准确、灵敏的定性定量分析。

In order to establish a method for the simultaneous content determination of donkey-derived peptide A1 and donkey-derived peptide A2 in Ejiao Yishou oral liquid by ultrafiltration centrifugal purification and liquid chromatography-mass spectrometry,trypsin hydrolysis,purification by 3kD ultrafiltration centrifuge tube,and liquid-mass spectrometry were used.The contents of the two characteristic peptides were determined simultaneously using a mass spectrometer(Waters Symmetry C18(150×2.1 mm,5μm)chromatographic column,mobile phase acetonitrile,0.1%formic acid gradient elution).It was found that the linearity of donkey-derived peptide A1 and donkey-derived peptide A2 was good(r≥0.9965),and the mean recoveries of the spiked samples were 101.13%and 88.54%with RSD of 4.19%and 6.63%respectively.This method can achieve rapid,accurate and sensitive qualitative and quantitative analysis of Chinese patent medicines containing Ejiao.
