LUCAT1在胃癌组织中的表达及对胃癌细胞增殖侵袭的影响

Expression of LUCAT1 in Gastric Cancer and Its Effect on Proliferation and Invasion of Gastric Cancer Cells

目的:探索肺癌相关转录本1(lung cancer transcript 1,LUCAT1)在胃癌组织中的表达及对胃癌细胞增殖侵袭的影响。方法:收集18例胃癌组织及配对癌旁正常组织,利用RT-qPCR检测LUCAT1的表达。利用RT-qPCR检测不同胃癌细胞株中LUCAT1表达,选取表达量最高的SGC7901细胞株进行相关实验。在SGC7901细胞中分别转染阴性对照siRNA、LUCAT1、siRNA空载体质粒及过表达质粒pLV-LUCAT1,采用RT-qPCR检测各组SGC7901细胞LUCAT1表达,MTT检测细胞增殖能力,Transwell实验观察细胞迁移和侵袭能力。结果:胃癌组织中LUCAT1表达量高于癌旁正常组织,差异有统计学意义(P<0.05)。si-LUCAT1组中LUCAT1表达较si-NC组降低,而pLV-LUCAT1组中LUCAT1表达较pLV-NC组增加,差异均有统计学意义(P<0.01)。si-LUCAT1组细胞增殖水平显著低于si-NC组,pLV-LUCAT1组细胞增殖水平显著高于pLV-NC组。si-LUCAT1组细胞的侵袭能力显著低于si-NC组,pLV-LUCAT1组细胞的侵袭能力显著高于pLV-NC组。结论:LUCAT1在胃癌组织中高表达,促进胃癌细胞的增殖和侵袭。

Objective:To explore the expression of LUCAT1 in gastric cancer and its effect on the proliferation and invasion of gastric cancer cells.Methods:18 cases of gastric cancer tissue were collected and adjacent normal tissues were paired,and RT-qPCR was used to detect the expression of LUCAT1.RT-qPCR was used to detect the expression of LUCAT1 in different gastric cancer cell lines,and the SGC7901 cell strain with the highest expression level was selected for related experiments.The negative control siRNA,LUCAT1,siRNA empty plasmid and overexpression plasmid pLV-LUCAT1 were transfected into SGC7901 cells,respectively.The expression of LUCAT1 in SGC7901 cells in each group was detected by RT-qPCR.MTT is used to detect cell proliferation changes.The migration and invasion of cells were observed by transwell experiment.Results:The expression of LUCAT1 in gastric cancer tissues was higher than that in the adjacent normal tissues.The expression of LUCAT1 in the si-LUCAT1 group decreased compared to the si-NC group,while the expression of LUCAT1 in the pLV-LUCAT1 group increased compared to the pLV-NC group,with statistical significance(P<0.01).The cell proliferation level in the si-LUCAT1 group was significantly lower than that in the si-NC group,while the cell proliferation level in the pLV-LUCAT1 group was significantly higher than that in the pLV-NC group.The invasion ability of cells in the si-LUCAT1 group was significantly lower than that in the si-NC group,while the invasion ability of cells in the pLV-LUCAT1 group was significantly higher than that in the pLV-NC group.Conclusion:LUCAT1 is highly expressed in gastric cancer and promotes the proliferation and invasion of gastric cancer cells.
