电针对神经病理性疼痛大鼠脊髓P2X7受体及其对IL-lβ表达的影响

Effects of electroacupuncture on P2X7R and its mediated IL-lβin spinal cord of neuropathic pain rat

摘要 目的:探究电针对神经病理性疼痛大鼠脊髓P2X7受体(P2X7R)及其介导的白细胞介素-1β(IL-lβ)的影响。方法:选择坐骨神经慢性挤压损伤(CCI)动物模型,将成年雄性SD大鼠36只,按照随机数字表法随机分为常规组、CCI组、电针组,各组12只,电针组于造模后第7日选取造模侧"足三里""阳陵泉"穴给予电针治疗,连续电针7 d,每日30 min,电流设置为1 mA、2/100 Hz,常规组只进行假手术,CCI组行造模,且两组大鼠只固定,不给予电针治疗。所有大鼠在造模前及造模后第3、7、10、14日时测量机械缩足阈值(MWT)和热缩足反应潜伏期(TWL)。造模后第14日后取脊髓L 4~6节段并采用RT-qPCR和蛋白印迹法检测各组大鼠脊髓P2X7R和IL-lβ mRNA和蛋白的表达水平。 结果:与常规组比较,CCI组大鼠MWT和TWL均明显降低,差异具有统计学意义( P均<0.05)。与CCI组比较,电针组大鼠MWT和TWL均明显升高,差异具有统计学意义( P均<0.05)。与常规组比较,CCI组大鼠脊髓P2X7R、IL-lβ mRNA和蛋白表达均明显升高,差异具有统计学意义( P均<0.05)。与CCI组比较,电针组大鼠脊髓P2X7R、IL-lβ mRNA和蛋白表达均明显下降,差异具有统计学意义( P均<0.05)。 结论:电针具有缓解CCI大鼠神经病理性疼痛的作用,其机制可能是通过抑制脊髓P2X7R及其介导的IL-lβ的表达来实现的。 Objective To investigate the effects of electroacupuncture(EA)on P2X7 receptor(P2X7R)and its mediated interleukin-1β(IL-lβ)in spinal cord of rats with neuropathic pain.Methods The animal model was chronic constriction injury of sciatic nerve(CCI).Thrity-six male Sprague-Dawley rats were divided into normal group,CCI group and EA group by random number table,12 rats in each group.In EA group,EA treatment was given at 7th day after the modelling and continuous for 7 days,at acupoints of Zusanli and Yanglingquan,with 1 mA,2/100 Hz,30 min/d.Only sham operation was performed in the conventional group,and modeling was performed in the CCI group,the rats were just fixed without EA treatment in the two groups.The mechanical withdrawal threshold(MWT)and the thermal withdrawal latency(TWL)were detected before CCI modelling and on 3,7,10,14th day after the modelling.At 14th day,the L4-6 spinal cord of all rats were harvested and observed by RT-qPCR and Western blot.Results Compared with the normal group,the MWT and TWL were significantly decreased in the CCI group(all P<0.05),but significantly increased in the EA group(all P<0.05).Compared with the normal group,the mRNA and protein of P2X7 and IL-lβin spinal cord were significantly increased in the CCI group(all P<0.05).Compared with the CCI group,the mRNA and protein of P2X7 and IL-lβin spinal cord were significantly decreased in the EA group(all P<0.05).Conclusion Electroacupuncture could relieve neuropathic pain in CCI rats model,and the mechanism may inhibit the expression of P2X7R and IL-lβin spinal cord.
