正交试验优化藜麦(Chenopodium quinoa)离体再生体系

Optimization of in Vitro Regeneration System of Quinoa(Chenopodium quinoa)by Orthogonal Test

摘要 为了优化藜麦离体再生体系的培养条件,实现藜麦离体高效快速繁殖。本试验选取材料‘忻藜1号’茎段和子叶为外植体,采用正交设计筛选出最适于藜麦愈伤组织分化的激素(6-苄氨基嘌呤,萘乙酸)、光强组合。结果显示:不同外植体在MS+6-BA 1.5 mg/L+NAA 0.4 mg/L培养基中出愈率最高,且茎段比子叶诱导率高,分别为77%和60%;在光强为4000 lx以上,MS+6-BA 1.5 mg/L+NAA 0.2 mg/L培养基中,不定芽分化率最高,可达96%。本试验的正交设计为藜麦快速繁殖和遗传转化提供技术支持。 In order to optimize the cultivation conditions of the quinoa regeneration system in vitro and realize the efficient and rapid propagation of quinoa in vitro.In this test,the stem segment and cotyledons of'Xinli No.1'were selected as explants.The orthogonal design was used to screen out the most suitable hormone(6-BA,NAA)and light intensity combination for quinoa callus differentiation.The results showed that different explants had the highest recovery rate inMS+6-BA 1.5 mg/L+NAA 0.4 mg/L medium,and the induction rate of stem segments was higher than that of cotyledons,77%and 60%;The adventitious bud differentiation rate was the highest in the medium with a light intensity of 4000 lx and MS+6-BA 1.5 mg/L+NAA 0.2 mg/L,up to 96%.The orthogonal design of this experiment provides technical support for the rapid propagation and genetic transformation of quinoa.
