摘要
目的研究基质金属蛋白酶(MMP)基因单核苷酸多态性与围绝经期女性卵巢功能减退的关联。方法采用PCR-RFLP技术对围绝经期女性群体的MMP-3 rs3025058和MMP-9 rs17576位点进行检测,评估其基因多态性与围绝经期卵巢功能减退的关联。结果卵巢功能衰竭组、减退组和正常组之间两位点的等位基因和基因型分布存在显著性差异(P<0.05),校正年龄因素后,携带基因型5A5A和AA的围绝经期女性发生卵巢功能减退的风险为6.702和7.936,携带基因型5A6A和AG的围绝经期女性发生卵巢功能减退的风险为2.886和5.315;显性模型下,携带等位基因5A和A的围绝经期女性发生卵巢功能减退的风险为2.886和5.315,卵巢功能衰竭组两位点的多态性与年龄相关,卵巢功能减退组rs17576多态性与年龄和FSH、E_(2)水平相关。结论MMP-3 rs3025058和MMP-9 rs17576单核苷酸多态性与围绝经期女性卵巢功能减退的发生存在明显关联,可作为围绝经期早期筛查的生物学标记。
Objective To study the association between single nucleotide polymorphism(SNP)of MMP gene and ovarian dysfunction in perimenopausal women.Methods PCR-RFLP was used to detect MMP-3 rs3025058 and MMP-9 rs17576 in perimenopausal women,and to evaluate the association between gene polymorphism and perimenopausal ovarian dysfunction.Results There were significant differ⁃ences in alleles and genotypes of the two loci between the ovarian failure group,the ovarian reverse group and the normal group(P<0.05),after adjusting for age factors,the risk of ovarian hypofunction was 6.702 and 7.936 in perimenopausal women with genotypes 5A5A and AA respectively,and 2.886 and 5.315 in perimenopausal women with genotypes 5A6A and AG respectively.Under the dominant model,the risk of ovarian hypofunction in perimenopausal women carrying alleles 5A and A was 2.886 and 5.315.The polymorphism of the two loci in the o⁃varian failure group was correlated with age,and the polymorphism of rs17576 in the ovarian hypofunction group was correlated with age and FSH and E_(2) levels.Conclusion MMP-3 rs3025058 and MMP-9 rs17576 single nucleotide polymorphisms are significantly associated with the occurrence of ovarian dysfunction in perimenopausal women,and can be used as biological markers for early screening of perimenopausal women.
