高效液相色谱法同时测定五味子中的4种木脂素成分

摘要 目的:建立高效液相色谱法(HPLC)同时测定五味子中五味子醇甲等4种木脂素成分的含量。方法:采用Sepax C18(250 mm×4.6 mm,5μm)色谱柱,甲醇-乙腈-水流动相梯度洗脱,流速1.0 mL/min,柱温30℃,进样量20μL,检测波长220 nm。结果:五味子醇甲、五味子酯甲、五味子甲素和五味子乙素分别在0.365~13.194 mg/g、0.388~14.853 mg/g、0.442~16.630 mg/g、0.313~11.882 mg/g浓度范围内呈良好的线性关系,回收率、精密度、稳定性等试验结果均符合方法学测定要求。12批不同产地五味子的4种木脂素成分含量有差异,RSD为0.76%~3.28%。结论:建立的高效液相色谱法可用于同时分析五味子中的4种木脂素成分。 Objective:To establish a high performance liquid chromatography(HPLC)method for simultaneous determination Schizandrol A and other three lignans in Schisandra Chinensis.Methods:HPLC separations were carried out on a Sepax C18 column(250 mm×4.6 mm,5μm)by gradient elution using methanol,acetonitrile and water as mobile phase.The flow rate was 1.0 mL/min,and the column temperature was 30°C.The injection volume was 20μL and the detection wavelength was 220 nm.Results:The linear relationships of Schizandrol A,Schisantherin A,Schizandrin A and Schizandrin B were good within the ranges of 0.36~13.20,0.39~14.86,0.44~16.63,and 0.32~11.88 mg/g,relatively.The results of recovery,precision,stability and so on all met the requirements of methodological determination.The content of the four lignans tested in 12 batches of Schisandra Chinensis samples from different producing areas was different,with the RSD of 0.76%~3.28%.Conclusin:The established method can be applied to determine the four lignans in Schisandra Chinensis simultaneously.
