ATM启动子甲基化与肿瘤相关性的研究进展

摘要 毛细血管扩张-共济失调症突变基因(ATM)是重要的抑癌基因和DNA修复基因,其参与细胞DNA损伤的识别、修复,细胞周期及凋亡调控等。DNA甲基化是表观遗传修饰的主要方式之一,ATM启动子异常甲基化可引起ATM表观沉默,导致细胞的肿瘤易感性及放射敏感性增加,研究ATM启动子甲基化可为精准放射治疗及放射治疗增敏提供新的思路。该文主要对ATM启动子甲基化与肿瘤的相关性研究进行综述,为深入研究ATM启动子甲基化对肿瘤早期诊断、肿瘤放射治疗增敏的作用提供依据。 Ataxia telangiectasia-mutated(ATM)gene is an important tumor suppressor gene and DNA repair gene,which involved in cell DNA damage recognition and repair,cell cycle and apoptosis regulation,ect.DNA methylation is one of the main ways of epigenetic modification.Abnormal methylation of ATM promoter can cause ATM apparent silence,which leads to the increase of tumor susceptibility and radiosensitivity of cells.Studying the methylation of ATM promoter can provide new ideas for precise radiotherapy and radiosensitization.In this paper,the correlation between methylation of ATM promoter and tumor was reviewed,in order to provide a basis for further study on the role of methylation of ATM promoter in early diagnosis and radiotherapy sensitization of tumor.
