基于生信分析探索PNPT1表达对非小细胞肺癌的影响 被引量:1

Exploration the effect of PNPT1 expression on non-small cell lung cancer based on bioinformatics analysis

摘要 目的探讨PNPT1的表达对非小细胞肺癌及肿瘤微环境的影响。方法基于TCGA数据集中的肺腺癌和肺鳞癌相关数据以及GTEx数据集中的健康人相关数据,使用GEPIA2数据库挖掘PNPT1在非小细胞肺癌组织与正常组织的表达差异,并通过绘制生存分析评估PNPT1表达量对非小细胞肺癌患者预后的影响。使用TIMER2.0数据库基于CIBERSORT算法进行PNPT1表达与免疫细胞浸润水平相关性预测,并使用GEPIA2数据库分析肿瘤组织中PNPT1与炎症和肿瘤杀伤相关因子编码基因的关联,以评价PNPT1表达水平对肿瘤微环境的影响。结果在非小细胞肺癌肿瘤组织中PNPT1表达增加(肺鳞癌:log2FC=0.976,P<0.01;肺腺癌:log2FC=0.331,P<0.01),且在不同分期间差异有统计学意义(F=3.17,P<0.05)。免疫细胞浸润相关性分析结果显示,PNPT1表达水平与非小细胞癌M1型巨噬细胞水平成正相关(肺腺癌:R=0.232,P<0.01;肺鳞癌:R=0.118,P<0.01),在肺腺癌组织中发现PNPT1与STAT1以及白细胞介素(IL)-6、CXCL8和IL-12A等炎症因子的编码基因呈正相关(R>0,P<0.05),同时与肿瘤杀伤因子IFNG(R=0.13,P<0.01)以及GZMB(R=0.16,P<0.01)间存在正相关。结论PNPT1在非小细胞肺癌中表达升高,PNPT1表达水平与非小细胞肺癌肿瘤免疫具有一定关联。 Objective To investigate the effect of PNPT1 expression on non-small cell lung cancer((NSCLC)and tumor microenvironment(TME).Methods Related data of lung adenocarcinoma(LUAD)and lung squamous cell carcinoma(LUSC)were extracted from TCGA data set,and the related data of healthy people were supplemented based on GTEx data set.GEPIA2 database was used to mine the expression difference of PNPT1 in NSCLC tissues and normal tissues.The influence of PNPT1 expression on the prognosis of NSCLC patients was evaluated by survival analysis.TIMER2.O database was used to predict the correlation between PNPT1 expression and immune cell infiltration level based on CIBERSORT algorithm.GEPIA2 database was used to analyze the association between PNPT1 and genes encoding inflammation and tumor killing related factors in tumor tissues,so as to evaluate the impact of PNPT1 expression level on TME.ResultsThe PNPT1 expression increased in NSCLC(LUSC:log2FC=0.976,P<0.01;LUAD:log_(2)FC=0.331,P<0.01)and showed a significant difference between different stages(F=3.17,P<0.05).The correlation analysis of immune cell infiltration showed that the expression level of PNPT1 was positively correlated with the level of M1 macrophages in NSCLC(LUAD:R=0.232,P<0.01;LUSC:R=0.118,P<0.01).PNPT1 was found to be positively correlated with coding genes of STATI and inflammatory factors interleukin(IL)-6,CXCL8 and IL-12A in lung adenocarcinoma tissues(R>0,P<0.05),and with tumor killing factors IFNG(R=0.13,P<0.01)and GZMB(R=0.16,P<0.01).Conclusion The expression of PNPT1 is increased in NSCLC and is related to the tumor immunity of NSCLC.
