基于化学发光法的凤头姜仔姜与老姜醇溶黄酮的抗氧化能力比较 被引量:2

Comparison of Antioxidant Capacity of Alcohol-Soluble Flavonoids Between Tender and Old Fengtou Ginger Based on Chemiluminescence Method

摘要 文章以凤头姜仔姜和老姜为实验材料,利用乙醇萃取反复沉淀的方法制备凤头姜仔姜、老姜醇溶黄酮,采用化学发光法构建了超氧阴离子自由基(O_(2)^(-)·)、羟自由基(·OH)、H_(2)O_(2)和对DNA损伤的保护作用4种抗氧化体系,分别测定凤头姜仔姜、老姜醇溶黄酮在4种不同抗氧化体系中的抗氧化能力。结果显示,凤头姜仔姜、老姜清除O_(2)^(-)·的半抑制浓度(half maximal inhibitory concentration,IC 50)分别为(26.7613±0.4168),(31.1083±0.3370)μg/mL;清除·OH的IC_(50)分别为(4.2763±0.8898),(14.3950±0.9696)μg/mL;清除H_(2)O_(2)的IC_(50)分别为(0.1723±0.0021),(0.1720±0.0044)μg/mL;对DNA损伤保护的IC_(50)分别为(6.8320±1.0206),(8.5470±0.4616)μg/mL。T检验发现凤头姜仔姜醇溶黄酮的抗氧化能力显著强于老姜,但在过氧化氢体系中,凤头姜仔姜的抗氧化能力与老姜无显著差异。 In this paper,with tender and old Fengtou ginger as the experimental material,the alcohol-soluble flavonoids of tender and old Fengtou ginger are prepared by ethanol extraction and repeated precipitation.Four antioxidation systems including superoxide anion free radical(O_(2)^(-)·),hydroxyl free radical(·OH),H_(2)O_(2) and the protection effect of DNA damage are established by chemiluminescence method.The antioxidant capacity of alcohol-soluble flavonoids of tender and old Fengtou ginger in the four different antioxidant systems is determined respectively.The results show that the half maximal inhibitory concentration(IC_(50))of tender and old Fengtou ginger on scavenging O_(2)^(-)·is(26.7613±0.4168),(31.1083±0.3370)μg/mL;the IC_(50) on scavenging·OH is(4.2763±0.8898),(14.3950±0.9696)μg/mL;the IC_(50) on scavenging H_(2)O_(2) is(0.1723±0.0021),(0.1720±0.0044)μg/mL;the IC_(50) on the protection of DNA damage is(6.8320±1.0206),(8.5470±0.4616)μg/mL.T test shows that the antioxidant capacity of tender Fengtou ginger alcohol-soluble flavonoids is significantly stronger than that of old ginger,but there is no significant difference between the antioxidant capacity of tender and old Fengtou ginger in hydrogen peroxide system.
