基于生物信息学方法分析B3GNT7基因在甲状腺癌组织中的表达水平及其对免疫细胞浸润的影响

摘要 目的基于生物信息学方法分析β-1,3-N-乙酰氨基葡萄糖转移酶7(B3GNT7)基因在甲状腺癌组织中的表达水平及其对免疫细胞浸润的影响。方法在肿瘤免疫评估资源数据库、GEPIA2数据库、UALCAN数据库中检索B3GNT7基因在甲状腺癌患者癌组织和癌旁组织中的表达情况,分析不同临床特征、病理分期甲状腺癌患者癌组织中B3GNT7基因的表达水平,采用多因素COX回归模型分析B3GNT7基因表达水平与甲状腺癌患者预后的关系,采用Spearman相关性模型分析B3GNT7基因表达水平与甲状腺癌组织中免疫细胞浸润丰度的相关性,以及B3GNT7基因表达水平与免疫细胞标志物、T淋巴细胞标志物、免疫检查点基因表达水平的相关性。结果B3GNT7基因在甲状腺癌患者癌组织中的表达高于癌旁组织(P<0.05)。不同病理分期的甲状腺癌患者癌组织中的B3GNT7基因表达水平具有差异性(P<0.05)。多因素COX分析结果显示,CD8^(+)T淋巴细胞、B3GNT7基因表达水平是甲状腺癌患者预后的影响因素(均P<0.05)。Spearman相关性分析结果显示,B3GNT7基因表达水平与B淋巴细胞、CD4^(+)T淋巴细胞、CD8^(+)T淋巴细胞、巨噬细胞、树突状细胞、中性粒细胞的浸润丰度,以及细胞毒性T淋巴细胞相关蛋白4(CTLA4)的表达水平密切相关(均P<0.05),还与多种免疫标志物的表达水平相关。结论B3GNT7基因在甲状腺癌组织中呈高表达,其表达水平与甲状腺癌患者的病理分期密切相关,B3GNT7基因与甲状腺癌组织中B淋巴细胞、CD4^(+)T淋巴细胞、CD8^(+)T淋巴细胞、巨噬细胞、树突状细胞、中性粒细胞的浸润丰度相关,且与CTLA4表达密切相关。 Objective To analyze the effect ofβ-1,3-N-acetylglucosaminyltransferase 7(B3GNT7)gene expression in thyroid carcinoma tissues and its on immune cell infiltration based on bioinformatics method.Methods The B3GNT7 gene expression in cancerous and paracancerous tissues of patients with thyroid carcinoma was retrieved from the databases of Tumor Immune Estimation Resource,GEPIA2,and UALCAN.The expression of B3GNT7 gene in cancerous tissues was analyzed in thyroid carcinoma patients with different clinicopathological features and pathological stages.The relation between B3GNT7 gene expression and prognosis of patients with thyroid carcinoma was analyzed by using multivariate COX regression model.The correlations of B3GNT7 gene expression with abundance of immune cell infiltration in thyroid carcinoma tissues,and with expressions of immunomarkers,T lymphocyte markers,and immune checkpoint genes were analyzed by employing the Spearman correlation model.Results The B3GNT7 gene expression in cancerous tissues was higher than that in paracancerous tissues of patients with thyroid carcinoma(P<0.05).There was difference in B3GNT7 gene expression in cancerous tissues between the thyroid carcinoma patients with different pathological stages(P<0.05).The results of multivariate COX analysis revealed that CD8^(+)T lymphocyte,B3GNT7 gene expression were the influencing factors for the prognosis of patients with thyroid carcinoma(all P<0.05).The results of Spearman correlation analysis implied that B3GNT7 gene expression closely correlated with infiltration abundance of B lymphocyte,CD4^(+)T lymphocyte,CD8^(+)T lymphocyte,macrophage,dendritic cell,and neutrophil,as well as with cytotoxic T-lymphocyte-associated protein 4(CTLA4)expression(all P<0.05);furthermore,B3GNT7 gene expression also correlated with expressions of multiple immunomarkers.Conclusion B3GNT7 gene is high expressed in thyroid carcinoma tissues,its expression closely correlates with pathological stages of patients with thyroid carcinoma,and correlates with infiltration abundance of B lymphocyte,CD4^(+)T lymphocyte,CD8^(+)T lymphocyte,macrophage,dendritic cell,and neutrophil,as well as with CTLA4 expression.
