摘要
目的优化葛枳汤的提取工艺。方法在单因素试验基础上,采用Box-Behnken响应面法设计试验,以提取时间、提取次数、加水量为影响因素,以芦丁、鸢尾苷、槲皮素、橙黄决明素含量为评价指标,采用信息熵法赋予权重系数,计算综合评分,通过响应面模型分析各影响因素间的交互作用,得出最佳提取工艺,并进行验证试验。结果优化的提取工艺为饮片浸泡0.5 h,加9倍量水,提取3次,每次分别提取90,45,45 min;验证试验结果显示,芦丁、鸢尾苷、槲皮素、橙黄决明素含量及综合评分的RSD分别为0.17%,2.79%,4.45%,0.52%,0.82%(n=3),与预测值的差异不超过1%。结论优化的提取工艺稳定、可靠,可为葛枳汤提取工艺的研究提供参考,也可为其新剂型的研发提供理论依据。
Objective To optimize the extraction process of Gezhi Decoction.Methods On the basis of the single factor experiment,Box-Behnken response surface methodology was used to design the experiment with the extraction time,extraction times,and additional amount of water as the investigation factors,and with the content of rutin,iridin,quercetin and aurantio-obtusin as the evaluation indexes.The weight coefficient was determined by the information entropy method,and the comprehensive score was calculated.The interaction among the influencing factors was analyzed by the response surface model,the optimal extraction process was obtained,and the validation test was carried out.Results The optimal extraction process was as follows:the decoction pieces were soaked for 0.5 h,added nine times the amount of water,extracted three times,and extracted for 90,45,and 45 min each time.The validation test results showed that the RSDs of rutin,iridin,quercetin and aurantio-obtusin content and comprehensive score were 0.17%,2.79%,4.45%,0.52%,and 0.82%(n=3),respectively,and the difference with the predicted value was not more than 1%.Conclusion The optimal extraction process is stable and reliable,which can provide a reference for the research of the extraction process of Gezhi Decoction and a theoretical basis for the development of its new dosage form.
