摘要
目的探讨PTPRD、PARD3B和TOMM7基因多态性与怀化地区人群高血压的相关性,为高血压的防治提供可靠的理论依据。方法采用病例对照研究方法,根据高血压诊断标准将研究人群分为高血压组与健康对照组,利用多重PCR-SNaPshot法对两组人群的PTPRD(rs649891、rs17584499)、PARD3B(rs849230)、TOMM7(rs2240727)4个基因位点进行基因分型,同时对两组人群进行问卷调查、体格测量和生理生化指标检测。结果单因素分析结果发现总胆固醇、甘油三酯、年龄、BMI、WHR、高脂高蛋白饮食、rs649891和rs17584499在两组中的分布差异均有统计学意义(P<0.05)。二分类logistic回归分析结果表明,年龄、BMI为超重或肥胖、中心性肥胖、高脂高蛋白饮食与高血压的发生呈正相关(P<0.05),rs649891的CT基因型与高血压的发生呈负相关(P<0.05)。结论PTPRD基因的rs649891位点与怀化地区人群高血压有关,其CT基因型可能是怀化地区人群高血压的保护因素,年龄、BMI、WHR、高脂高蛋白饮食为怀化地区人群高血压的危险因素。
Objective To explore the correlation between the polymorphism of PTPRD,PARD3B and TOMM7 gene and hypertension in Huaihua area,and to provide reliable theoretical basis for the prevention and treatment of hypertension.Methods A case-control study was conducted to divide the populations into hypertension group and healthy control group according to the diagnostic criteria of hypertension.The PTPRD(rs649891,rs17584499),PARD3B(rs849230),and TOMM7(rs2240727)gene loci of the two groups were genotyped by multiple PCR-SNaPshot method.At the same time,questionnaire survey,physical measurement,and physiological and biochemical indicators were conducted for the two groups.Results The results of univariate analysis showed that the distribution differences of total cholesterol,triglyceride,age,BMI,WHR,high-fat and high-protein diet,rs649891 and rs17584499 in the two groups were statistically significant(P<0.05).The results of binary logistic regression analysis showed that age,BMI as overweight or obesity,central obesity,high-fat and high-protein food were positively correlated with the occurrence of hypertension(P<0.05),and the CT genotype of rs649891 was negatively correlated with the occurrence of hypertension(P<0.05).Conclusion The rs649891 locus of PTPRD gene is associated with hypertension in Huaihua area,and its CT genotype may be a protective factor for hypertension in Huaihua area,while age,BMI,WHR,high fat and high protein diet are risk factors for hypertension in Huaihua area.
