无硝酸铵的改良MS培养基对马铃薯组培苗快繁的影响

摘要 马铃薯组培苗快繁生产中,因硝酸铵受管控,迫切需要找到硝酸铵的良好替代物。以2个马铃薯品种为试验材料,采用(NH_(4))_(2)SO_(4)、KNO_(3)替代NH_(4)NO_(3)的改良MS培养基配方,研究其对马铃薯组培苗生长的影响。结果表明:在(NH_(4))_(2)SO_(4)单独替代NH_(4)NO_(3)的MS培养基中,2个马铃薯品种的组培苗均表现茎叶、根鲜重显著减少;在KNO_(3)单独替代NH_(4)NO_(3)的MS培养基中,2个马铃薯品种的组培苗均出现不同程度黄叶、矮小现象;而在(NH_(4))_(2)SO_(4)、KNO_(3)混合替代NH_(4)NO_(3)的MS培养基中,马铃薯组培苗均能生长正常,其组培苗快繁适宜的硝酸铵替代方案为1.36 g·L^(-1)(NH_(4))_(2)SO_(4)+1.04~2.08 g·L^(-1)KNO_(3)。 In the production of rapid propagation of potato tissue culture seedlings,there is an urgent need to find a good alternative to ammonium nitrate due to the controlled use of ammonium nitrate.In this study,two potato varieties were used as materials to study the effects of different modified MS formulas in which(NH_(4))_(2)SO_(4)and KNO_(3)were used as substitute to NH_(4)NO_(3) on growth of potato tissue culture seedlings.The results showed that fresh weight of stem,leaf and root of seedlings significantly decreased in MS medium with(NH_(4))_(2)SO_(4)as substitute to NH_(4)NO_(3);seedlings showed yellow leaves and dwarf plant in MS medium with KNO_(3) as substitute to NH_(4)NO_(3);seedlings grew up regularly in MS medium with(NH_(4))_(2)SO_(4) and KNO_(3) as substitute to NH_(4)NO_(3).It is a suitable plan that 1.36 g·L^(-1)(NH_(4))_(2)SO_(4)and 1.04~2.08 g·L^(-1)KNO_(3)were used as substitute to NH_(4)NO_(3).
