鹅掌疯醋剂对抗白色念珠菌及其早期生物膜的体外实验研究

Effect of Ezhangfeng Vinegar on Candida albicans and Its Early Biofilm:An in Vitro Experimental Study

目的 分析鹅掌疯醋剂对白色念珠菌及早期生物膜的作用及其机制。方法 通过微量稀释法测定鹅掌疯醋剂对白色念珠菌的最小抑菌浓度(minimum inhibitory concentration,MIC),通过酶标仪测得其对菌体渗透性的影响,使用扫描电子显微镜和透射电子显微镜观察其对菌体超微结构的影响,并且通过CCK-8法和Spider平板法测定其对白色念珠菌生物被膜形成早期黏附性和芽管形成的影响。结果 鹅掌疯醋剂对白色念珠菌的MIC为药物原浓度的1/16,经过鹅掌疯醋剂处理后白色念珠菌细胞外紫外物质吸收增加,念珠菌超微结构被破坏,早期生物膜的黏附率和芽管形成受到抑制。结论 鹅掌疯醋剂有一定的抗真菌及抑制早期生物膜能力,其作用机制可能与改变菌体渗透性,破坏菌体结构,抑制早期生物膜的黏附率和芽管形成有关。

Objective To investigate the effect of Ezhangfeng vinegar on Candida albicans and its early biofilm and related mechanism. Methods The microdilution method was used to determine the minimum inhibitory concentration (MIC) of Ezhangfeng vinegar against Candida albicans , and a microplate reader was used to observe its effect on fungal permeability. Scanning electron microscopy and transmission electron microscopy were used to observe the influence of Ezhangfeng vinegar on fungal ultrastructure, and CCK-8 assay and the Spider plate method were used to observe its influence on adhesion in the early stage of Candida albicans biofilm formation and the formation of germ tube. Results The MIC of Ezhangfeng vinegar against Candida albicans was 1/16 of the original concentration of the drug, and after Ezhangfeng vinegar treatment, there was an increase in the uptake of extracellular ultraviolet substance, destruction of Candida ultrastructure, and inhibition of early biofilm adhesion rate and germ tube formation. Conclusion Ezhangfeng vinegar has a certain antifungal effect and can inhibit early biofilm formation, possibly by altering fungal permeability, destroying fungal structure, and inhibiting the adhesion rate of early biofilm and the formation of germ tube.
