摘要
本试验旨在优化刺三加根总黄酮的最佳提取条件,并对其抗氧化性能进行测定。先通过单因素试验优化提取拟定的工艺参数,并进一步用响应面法优化得到二次多项数学模型。结果表明:最佳提取工艺为乙醇浓度35%、料液比1:40 g/mL、超声温度25℃、时间35 min,此条件下提取率为7.59%,相当于总抗氧化能力为3.361 mmol/L(FeSO_(4)当量值),而0.4 mg/mL提取物的DPPH自由基清除率可达到89.4%。本试验易操作、提取效率高,可用于刺三加根中的总黄酮提取,且提取液中的总黄酮可展现出好的抗氧化性能,因此,可为其资源开发提供依据。
The optimal extraction conditions and antioxidant properties of total flavonoids from Acanthopanax trifoliatus root were optimized by experiments.The proposed process parameters of extraction were optimized by single factor experiment,and the quadratic multiphase mathematical model was obtained further by optimization from response surface methodology.The results showed that the extraction process parameters were 35%ethanol,1:40 mL/g solid-liquid ratio,25℃ultrasonic temperature combined with 35 min.The corresponding extraction rate was 7.59%,corresponding to 3.361 mmol/L(FeSO_(4) equivalent value)of the total antioxidant capacity,and then,the DPPH free radical scavenging rate of the 0.4 mg/mL extract could reach 89.4%.This experiment was easy to operate,and high in extraction efficiency,which could be used for the total flavonoids’extraction from Acanthopanax trifoliatus roots.Andthen,the total flavonoids of the extract could exhibit good antioxidant performance,so a basis for its resource development could be provided.
