摘要
为揭示固始鸡胸肌脂肪酸组成特征及其关键调控基因,试验采用气相色谱-质谱联用仪(GC-MS)技术分析了14、22和30周龄固始鸡胸肌组织的脂肪酸组成,通过权重基因共表达网络分析鉴定了主要脂肪酸含量相关的核心基因。结果显示:三个发育阶段胸肌组织中均检测到49种脂肪酸,且多数脂肪酸含量在不同发育阶段间差异不显著(P>0.05);胸肌组织中优势脂肪酸为C16∶0、C18∶0、C18∶1N12、C18∶1N9C、C20∶4N6、C18∶1N7、C18∶2N6和C14∶1T,含量占总脂肪酸含量的86%以上。另外,共鉴定到blue、brown、purple、lightyellow、grey60和red共6个特异性转录模块,从中筛选到104个与C14∶0、C16∶0、C17∶0、C18∶1N12、C18∶2N6、C18∶3N6、C20∶1、C20∶2和C20∶3N6等脂肪酸含量相关的核心基因,功能涉及脂肪酸代谢相关的许多生物学过程和信号通路。结果表明,固始鸡胸肌脂肪酸含量在不同生长时期存在动态变化且为微效多基因共同调控的复杂数量性状。
In order to reveal the characteristics of fatty acid composition and its key regulatory genes in Gushi chicken breast muscle,the fatty acid composition of in breast muscle in Gushi chicken at the age of 14,22,and 30 weeks was detected by the GC-MS technology,and hub genes related to the content of major fatty acids were identified by weighted gene coexpression network analysis.The results showed that 49 fatty acids were detected in all three stages of breast muscle tissue,and the contents of most fatty acids were not significantly different between different development stages(P>0.05).The content of fatty acids,such as C16∶0,C18∶0,C18∶1N12,C18∶1N9C,C20∶4N6,C18∶1N7,C18∶2N6 and C14∶1T,had a significant advantage in breast muscle tissue,accounting for more than 86%of the total fatty acid content.In addition,six specific transcriptional modules were identified,which included the blue,brown,purple,lightyellow,grey60 and red modules.A total of 104 hub genes related to the content of fatty acid C14∶0,C16∶0,C17∶0,C18∶1N12,C18∶2N6,C18∶3N6,C20∶1,C20∶2 and C20∶3N6 were screened from these specific transcriptional modules.These hub genes were involved in many biological processes and signaling pathways relat⁃ed to fatty acid metabolism.The results indicated that.The results indicated that the fatty acid composition in Gushi chicken breast muscle varies dynamically during different growth periods and is a complex quantitative trait regulated by a combination of micro-effective genes.
