翻译后修饰对结核分枝杆菌耐药性调控机制的研究

摘要 结核病是由结核分枝杆菌引发的慢性、致死性传染病,耐药株的出现增加了其防治难度。早期针对耐药机制的研究多围绕其发生机制和药物靶点展开。近年来,越来越多的研究显示结核分枝杆菌中大量蛋白质存在翻译后修饰,这些蛋白调控生理生化代谢等重要生命过程,并与耐药性产生存在一定关系。本文系统性地总结了结核分枝杆菌中蛋白质翻译后修饰与其获得性耐药之间的关系,阐述了常见类型翻译后修饰对结核耐药的调控机制;为进一步深入研究翻译后修饰的生物学意义、探究抗结核药物的作用机理、降低结核病对社会的危害以及药物作用靶点研发、治疗方案调整等提供新的思路。 Tuberculosis is a chronic and lethal infectious disease caused by Mycobacterium tuberculosis(MTB).The emergence of drug-resistant strains has hindered tuberculosis prevention and treatment.Previous studies in this area have focused primarily on pathogenesis and drug targets.Post-translational modifications(PTMs)have been discovered in many MTB proteins.These modified proteins regulate the physiological and biochemical metabolism of MTB and may influence tuberculosis drug resistance.Hence,this article systematically summarizes the relationship between PTMs and the acquisition of drug resistance by MTB,and describes the regulatory mechanisms of common types of PTMs in tuberculosis drug resistance.This work provides insights into the biological importance of PTMs and the mechanisms of antituberculosis drugs for further study,which may contribute to decreasing the societal harm of tuberculosis,and aid in the discovery of new drug targets and therapeutic regimens.
