乌达巴拉-8味散抑制博来霉素致小鼠特发性肺纤维化的研究

Study on the Inhibition of Wudabala-8 Flavor Powder on Bleomycin-induced Idiopathic Pulmonary Fibrosis in Mice

摘要 目的:观察蒙药乌达巴拉-8味散对博来霉素致小鼠肺纤维化的影响。方法:选用C57BL/6小鼠30只,分为空白组、模型组、乌达巴拉-8味散组,每组10只。除空白组外,其余两组小鼠用博来霉素(5 mg·kg^(-1)),单次气管内注射的方法造模,建模2 d开始灌胃给药,每日1次,空白组给予CMC-Na罐胃,模型组给予CMCNa灌胃,给药组用乌达巴拉-8味散混悬液灌胃(1.5 g·kg^(-1)),每组灌胃体积均为20 mL·kg^(-1),每周称重1次并调整给药量,连续给药28 d后,眼球摘除法处死小鼠,取肺组织,采用苏木素-伊红(HE)、Masson染色评价肺组织炎症和纤维化程度,使用免疫组化法检测肺组织I型胶原(collagen I)、Ⅲ型胶原(collagenⅢ)蛋白的表达。结果:空白组小鼠肺部组织未见明显异常;模型组小鼠肺部组织可见肺泡结构破坏明显,部分肺间质被纤维细胞替代,并伴有炎性细胞浸润,明显有胶原沉积;乌达巴拉-8味散组小鼠肺组织肺泡结构破坏明显减少,炎症细胞和胶原沉积减少;与空白组比较,模型组小鼠蓝色区域面明显增大,差异有统计学意义(P<0.05);与模型组比较,给药组小鼠肺部组织蓝色区域面明显降低,差异有统计学意义(P<0.05);与空白组比较,模型组小鼠肺组织I型、Ⅲ型胶原蛋白的表达量都呈强阳性;与模型组比较,给药组小鼠肺组织I型胶原蛋白、Ⅲ型胶原蛋白表达强度降低。结论:蒙药乌达巴拉-8味散能有效减轻博来霉素致小鼠特发性肺纤维化形成并能减少肺组织I型胶原蛋白、Ⅲ型胶原蛋白的表达。 Objective:To investigate the effect of Wudabala-8 flavor powder on bleomycin-induced pulmonary fi⁃brosis in mice.Methods:Thirty C57BL/6 mice were selected and divided into a blank group,a model group and a Wudabala-8 flavor powder group,10 mice in each group.Except the blank group,the other two groups of mice were molded by single intratracheal injection of bleomycin(5 mg·kg^(-1)),and the drug was given by gavage 2 days after modeling,once a day.The blank group and the model group were given CMC-Na by gavage,the drug group was given Wudabala-8 flavor powder suspension(1.5 g·kg^(-1))by gavage,and the gavage volume of each group was 20 mL·kg^(-1).The mice were weighed once a week and the administration dose was adjusted.After con⁃tinuous administration for 28 days,the mice were sacrificed by eye extraction,and the lung tissue was taken out.Hematoxylin-eosin(HE)and Masson stains were used to evaluate the degree of inflammation and fibrosis of lung tissue,and immunohistochemistry was used to detect the expression of collagenⅠand collagenⅢproteins in lung tissue.Results:There were no obvious abnormalities in the lung tissues of mice in the blank group.The lung tissue of mice in the model group showed obvious destruction of alveolar structure,part of the lung stroma was replaced by fibroblasts,accompanied by inflammatory cell infiltration and obvious collagen deposition.The destruction of alveolar structure in lung tissue,inflammation cells and collagen deposition were significantly re⁃duced in the Wudabala-8 flavor powder group.Compared with the blank group,the blue area of mice lungs in the model group was significantly increased,and the difference was statistically significant(P<0.05).Compared with the model group,the blue area of mice lungs in the drug group was significantly reduced,and the difference was statistically significant(P<0.05).Compared with the blank group,the expression of typeⅠand typeⅢcolla⁃gen in lung tissues of mice in the model group was strongly positive.Compared with the model group,the expres⁃sion intensity of typeⅠcollagen and typeⅢcollagen in mice of the drug group was reduced.Conclusion:Wuda⁃bala-8 flavor powder can effectively reduce the formation of idiopathic pulmonary fibrosis in mice caused by bleomycin and reduce the expression of typeⅠcollagen and typeⅢcollagen in lung tissue.
