摘要
目的采用超高效液相色谱法(UPLC)建立穿心莲药材特征图谱及4个成分的一测多评方法。方法通过对12批穿心莲药材特征图谱的测定,建立穿心莲药材特征图谱共有模式,并进行相似度评价,经高分辨质谱分析和对照品比对,对特征峰进行指认。以穿心莲内酯为参照,建立新穿心莲内酯、14-去氧穿心莲内酯和脱水穿心莲内酯的一测多评法,并与外标法测定结果比较,判断方法的准确性。结果穿心莲药材特征图谱相似度为0.905~0.995,共确定12个特征峰,指认出其中7个。一测多评法与外标法测定结果的相对偏差在5.0%以内,表明该方法准确性良好。结论特征图谱结合一测多评法能够有效评价穿心莲药材的质量,该方法准确、可靠,系统适用性良好。
Objective To establish the characteristic chromatogram of Andrographis Herba and quantitative analysis of multi-components by single marker(QAMS)of four components by ultra performance liquid chromatography(UPLC).Methods Through the determination of the specific chromatograms of 12 batches of Andrographis Herba,the common pattern of the specific chromatogram of Andrographis Herba was established and the similarity was evaluated.The characteristic peaks were identified by high resolution mass spectrometry analysis and comparison with reference materials.With andrographolide as a reference,quantitative analysis of QAMS was established for new neoandrographolide,14 deoxyandrographolide and dehydrated andrographolide and the results were compared with the external standard method(ESM)to determine the accuracy of the method.Results The similarity of the specific chromatograms of Andrographis Herba was 0.905–0.995,and 7 of the 12 characteristic peaks were identified.The relative deviation between QAMS and ESM was within 5.0%,indicating that the accuracy of the method was good.Conclusion The specific chromatogram combined with QAMS can effectively evaluate the quality of Andrographis Herba,and the method is accurate,reliable and has good system applicability.
