急性缺血性脑卒中患者血清HDAC3和SMAD3水平变化与神经功能缺损程度的相关性分析 被引量:2

摘要 目的 探索急性缺血性脑卒中患者血清组蛋白去乙酰化酶3(HDAC3)和信号转导分子3(SMAD3)水平变化与神经功能缺损程度的相关性。方法 选取2021年6月-2022年6月在我院确诊为AIS的患者104例即为AIS组,根据美国国立卫生研究院卒中量表(NIHSS)对其神经功能损伤程度进行分类,其中轻度组32例,中度组37例,重度组35例。同期在我院体检中心体检健康的志愿者98例为对照组。采用酶联免疫吸附法(ELISA)检测血清中HDAC3、SMAD3、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)表达水平。采用Pearson法分析血清HDAC3和SMAD3表达的相关性;Spearman相关性分析血清中HDAC3、SMAD3表达与NIHSS评分、神经功能缺损程度的相关性。结果 与对照组相比,AIS组TG、TC、LDL-C、FIB、TNF-α、IL-1β、IL-6水平显著升高,HDL-C水平显著降低(P<0.05)。与对照组相比,AIS组HDAC3和SMAD3水平均显著升高(P<0.05)。重度组HDAC3和SMAD3的表达水平显著高于中度组及轻度组,中度组显著高于轻度组(P<0.05)。Pearson相关性分析显示,AIS患者血清中HDAC3和SMAD3表达水平呈正相关关系(P<0.05)。Spearman相关性分析显示,血清中HDAC3、SMAD3表达与NIHSS评分呈正相关关系(r=0.339,P<0.001;r=0.563,P<0.001)。血清HDAC3、SMAD3表达水平与神经功能缺损程度呈正相关关系(r=0.698,P<0.001;r=0.572,P<0.001)。结论 AIS患者血清中HDAC3和SMAD3表达水平异常升高,两者与神经功能缺损程度有关。 Objective To explore the correlation between serum levels of histone deacetylase 3(HDAC3)and the signal molecule SMAD family member 3(SMAD3)and the degree of neurological deficit in patients with acute ischemic stroke(AIS).Methods From June 2021 to June 2022,104 patients diagnosed with AIS in our hospital were selected.Using the National Institutes of Health Stroke Scale(NIHSS),they were divided based on the degree of neurological impairment into mild group(32 cases),moderate group(37 cases),and severe group(35 cases).At the same time,98 healthy volunteers from health examination in our hospital were included as the control group.The expression levels of HDAC3,SMAD3,tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)in serum were measured by using enzyme-linked immunosorbent assay.Pearson correlation analysis was used to analyze the correlation between the expression of HDAC3 and SMAD3.Spearman correlation analysis was used to analyze the correlation between the expression of HDAC3 and SMAD3 and NIHSS score and the degree of neurological deficit.Results Compared with the control group,the AIS group showed significantly higher levels of triglyceride,total cholesterol,low-density lipoprotein cholesterol,fibrinogen,TNF-α,IL-1β,and IL-6 and a significantly lower level of HDL-C(all P<0.05).HDAC3 and SMAD3 levels were significantly higher in the AIS group than in the control group(both P<0.05).HDAC3 and SMAD3 levels in the severe group were significantly higher than those in the moderate and mild groups,and those in the moderate group were significantly higher than those in the mild group(all P<0.05).The Pearson correlation analysis showed that the serum expression levels of HDAC3 and SMAD3 in the AIS patients were positively correlated(P<0.05).The Spearman correlation analysis showed positive correlations between the expression of HDAC3 and SMAD3 and NIHSS score(r=0.339,0.563;both P<0.001)and the degree of neurological impairment(r=0.698,0.572;both P<0.001).Conclusion Serum HDAC3 and SMAD3 levels in patients with AIS were abnormally increased,and both were related to the degree of neurological deficit.
