益肾通络方对多重损伤性大鼠薄型子宫内膜IL-2,IL-6表达的影响 被引量:2

Effect of Yishen-Tongluo Prescription on the expression of IL-2 and IL-6 in thin endometrium with multiple injuries of rat model

摘要 目的 探讨益肾通络方对多重损伤性大鼠子宫内膜IL-2、IL-6表达的影响。方法 采用SD雌性大鼠,建立肾虚血瘀证薄型子宫内膜模型。按随机数字表法分为空白组、模型组、补佳乐组、益肾通络方低、中、高剂量组。采用HE染色光镜下观察各组子宫内膜形态学变化,同时采用R T-PCR和Western blot法分别检测各组子宫内膜IL-2、IL-6 mRNA表达及蛋白水平。结果 与空白组比较,模型组大鼠子宫内膜明显变薄(P<0.01),IL-2、IL-6 mRNA表达及蛋白水平均明显升高(P<0.05);与模型组比较,益肾通络高剂量组子宫内膜明显增厚(P<0.01),IL-2、IL-6 mRNA表达及蛋白水平均明显降低(P<0.05)。结论 益肾通络方可通过下调子宫内膜IL-2、IL-6表达,降低子宫内膜局部的炎症反应,从而改善子宫内膜容受性。 Objective To explore the effect of Yishen Tongluo Prescription on the expression of IL-2 and IL-6 in endometrium with multiple injuries of rats.Methods The thin endometrium model of kidney deficiency and blood stasis was established with female SD rats.The rats were divided into a blank group,a model group,a progynova group,low-dose,medium-dose and high-dose Yishen Tongluo Prescription groups.HE staining was used to observe the morphological changes of the endometrium.The expressions of IL-2 and IL-6 mRNA in rat endometrium were quantitatively analyzed by RT-PCR.The expressions of IL-2 and IL-6 protein in the endometrium of rats in each group were detected by Western-blot.Results Compared with the blank group,the endometrium thickness of the model group was significantly thinner(P<0.05).Compared with the model group,the endometrium of the high-dose Yishen Tongluo Prescription group was significantly thickened(P<0.01),and the mRNA and protein expression levels ofIL-2 and IL-6 were significantly decreased(P<0.05).Conclusion Yishen Tongluo Prescription can reduce the local inflammatory response of endometrium and improve the endometrial receptivity by inhibiting the expression of IL-2 and IL-6 in endometrium of rats with in endometrium of rats with multiple injuries.
