摘要
目的:研究真武汤对阿霉素诱导慢性心力衰竭大鼠心肌细胞凋亡因子的影响。方法:将24只斯泼累格·多雷(SD)大鼠分为空白组、模型组、美托洛尔组、真武汤组,每组6只。利用腹腔注射阿霉素建立慢性心力衰竭模型。给药7周后,利用彩超仪检测心功能左心室射血分数(LVEF)、左心室短轴缩短率(LVFS);利用酶联免疫吸附试验(ELISA)法检测心肌组织B细胞淋巴瘤-2蛋白(Bcl-2)、Bcl-2相关X蛋白(Bax)、Bcl-2的浓度改变;蛋白免疫印迹(Western Blotting)法观察心肌组织Bax、Bcl-2、基质金属蛋白酶-9(MMP-9)蛋白含量。结果:与空白组比较,模型组LVEF、LVFS降低,差异有统计学意义(均P<0.01)。与模型组比较,真武汤组LVEF、LVFS升高(均P<0.01),BAX的浓度和蛋白表达显著降低(均P<0.01),Bcl-2的浓度及蛋白表达显著上升(均P<0.01),MMP-9的蛋白表达降低(P<0.01)。结论:真武汤能有效调控大鼠心肌细胞凋亡因子,进而改善大鼠心功能。
Objective:To study the effect of Zhenwu Decoction on the apoptosis of cardiomyocytes in the rat model of chronic heart failure induced by adriamycin.Methods:Twenty-four SD rats were assigned into a blank group,a model group,a metoprolol group,and a Zhenwu Decoction group.Chronic heart failure model was established by intraperitoneal injection of adriamycin.After 7 weeks of drug administration,the cardiac function indicators[left ventricular ejection fraction(LVEF)and left ventricular fraction shortening(LVFS)]were examined by a color doppler ultrasound instrument.Enzyme-linked immunosorbent assay was employed to measure the levels of B-cell lymphoma-2(Bcl-2)and Bcl-2-associated X protein(Bax)in the myocardial tissue.Western blotting was employed to determine the protein levels of Bax,Bcl-2,and matrix metalloproteinase-9(MMP-9)in the myocardial tissue.Results:Compared with the blank group,the modeling lowered the LVEF and LVFS(P<0.01).Compared with the model group,Zhenwu Decoction elevated LVEF and LVFS(P<0.01),lowered the serum level of Bax and down-regulated the protein level of Bax(P<0.01),elevated the serum level of Bcl-2 and up-regulated the protein level of Bcl-2(P<0.01),and down-regulated the protein level of MMP-9(P<0.01).Conclusion:Zhenwu Decoction can regulate the apoptosis factors of cardiomyocytes and thus improve the cardiac function of rats.
