罗伊氏乳杆菌对大鼠不同类型腹泻的影响 被引量:2

摘要 为研究罗伊氏乳杆菌的益生作用,本试验研究了蓝狐源罗伊氏乳杆菌ZJF036对葡聚糖硫酸钠(DSS)、沙门氏菌、庆大霉素(GEN)+头孢拉定(CE)诱导的腹泻的预防及治疗作用。本试验选取了80只2~4周龄SD大鼠,随机分为10组每组8只。整个试验周期21 d,正常对照组灌胃无菌生理盐水,DSS对照组只饮用2%DSS溶液(62.5 g/L),沙门氏菌对照组只灌胃沙门氏菌菌悬液(1×10^(8)CFU/mL),GEN+CE对照组只饮用GEN+CE溶液,DSS预防组、沙门氏菌预防组和GEN+CE预防组先灌胃罗伊氏乳杆菌后分别饮用DSS溶液、沙门氏菌菌悬液和GEN+CE溶液,DSS治疗组、沙门氏菌治疗组和GEN+CE治疗组分别先饮用DSS溶液、沙门氏菌菌悬液和GEN+CE溶液后灌胃罗伊氏乳杆菌菌悬液。试验结束计算大鼠体重总增重,测定各组大鼠血清中白细胞介素2(IL-2)、白细胞介素4(IL-4)、白细胞介素10(IL-10)、白细胞介素12(IL-12)和肿瘤坏死因子α(TNF-α)的水平。试验结果显示:与DSS对照组相比,DSS预防组大鼠总增重显著增加(P<0.05);与沙门氏菌对照组相比沙门氏菌治疗组与预防组大鼠的总增重显著增加(P<0.05);与GEN+CE对照组相比,GEN+CE预防组与治疗组总增重没有显著差异。与DSS对照组相比,DSS预防组大鼠血清中IL-2、IL-4、TNF-α的水平显著降低(P<0.05);DSS治疗组IL-12、TNF-α的水平显著降低(P<0.05);与沙门氏菌对照组相比,沙门氏菌治疗组大鼠血清中IL-2、IL-4、IL-12和TNF-α的水平显著降低(P<0.05),沙门氏菌预防组IL-12的水平显著降低(P<0.05);与GEN+CE对照组相比,GEN+CE治疗组与预防组IL-2、IL-12的水平显著降低(P<0.05),GEN+CE治疗组IL-10的水平显著提高(P<0.05)。由试验结果可知,灌服罗伊氏乳杆菌可改善沙门氏菌感染和DSS诱导的大鼠体重减轻情况,并且对各组大鼠血清中抗炎细胞因子白细胞介素4、白细胞介素10和促炎细胞因子白细胞介素2、白细胞介素12以及肿瘤坏死因子α有一定的调节作用。研究表明,罗伊氏乳杆菌ZJF036可改善这3种类型诱导的大鼠腹泻症状,缓解肠道炎症,通过调节细胞因子来维持肠道免疫稳态。 To study the probiotic effect of Lactobacillus reuteri,the preventive and treatment effects of Lactobacillus reuteri ZJF036 from fox origin on dextran sodium sulfate(DSS),Salmonella,gentamicin+cefradine induced diarrhea.In this experiment,80 SD rats aged 2-4 weeks were selected and randomly divided into 10 groups with 8 rats in each group.The normal control group was gavaged with sterile saline,the DSS control group only drank DSS solution,the Salmonella control group only gavaged Salmonella suspension,the GEN+CE control group only drank GEN+CE solution,the DSS prevention group,the Salmonella prevention group,and the GEN+CE prevention group were given first Lactobacillus reuteri respectively drank DSS solution,Salmonella bacteria suspension,GEN+CE solution,DSS treatment group,Salmonella treatment group,GEN+CE treatment group respectively drank DSS solution,Salmonella bacteria suspension,GEN+CE solution and then gavage Lactobacillus bacteria suspension.At the end of the test,the total weight gain of the rats was calculated,and the serumlevels of interleukins 2,interleukins 4,interleukins 10and interleukins 12(IL-2,IL-4,IL-10,IL-12),tumor necrosis factor α(TNF-α levels.The test results showed:①Compared with the DSS control group,the DSS prevention group significantly increased the total weight gain of the rats(P<0.05);Compared with the Salmonella control group,the Salmonella treatment group and the prevention group significantly increased the total weight gain of the rats(P<0.05).Compared with the GEN+CE control group,there was no significant difference in total weight gain between the GEN+CE prevention group and the treatment group.②Compared with the DSS control group,the DSS prevention group significantly decreased the serumlevels of IL-2,IL-4 and TNF-α(P<0.05);The DSS treatment group significantly decreased the levels of IL-12 and TNF-α compared with the Salmonella control group,the Salmonella treatment group significantly decreased the levels of IL-2,IL-4,IL-12 and TNF-α in the serumof the rats(P<0.05).Group significantly decreased the level of IL-12(P<0.05).Compared with the GEN+CEcontrol group,the GEN+CEtreatment group and the prevention group significantly decreased the levels of IL-2 and IL-12(P<0.05),and the GEN+CE treatment group significantly increased the IL-10 level(P<0.05).Fromthe test results,it can be seen that the administration of Lactobacillus reuteri can improve the weight loss of rats induced by Salmonella infection and DSS,and the anti-inflammatory cytokines interleukin 4,Interleukins 10 and pro-inflammatory cytokines leukocytes in the serum of rats in each group.Interleukins 2,Interleukins 12 and tumor necrosis factor have certain regulatory effects.It can be seen that Lactobacillus reuteri can improve the abdominal symptoms induced by these three types in rats,reduce intestinal inflammation,and maintain intestinal immune homeostasis.
