富硒小球藻提取物对小鼠急性肝损伤的预防和治疗作用

摘要 目的探究富硒小球藻提取物(含硒量3069 mg/kg)对小鼠急性肝损伤的预防和治疗作用。方法42只雄性C57小鼠随机分为:对照组、模型组、联苯双酯药物组、无硒组、低硒组、中硒组、高硒组,每组6只。预防试验中对照组和模型组每日灌胃20 mL/kg蒸馏水,药物组灌胃0.200 g/kg联苯双酯滴丸,补硒组分别为富硒小球藻提取物0.016、0.033、0.065 g/kg、无硒组为无硒破壁小球藻提取物0.065 g/kg,1次/d,连续7 d,末次灌胃1 h后,对照组腹腔注射大豆油,其余各组腹腔注射10%CCl 4大豆油溶液建立肝损伤模型,注射剂量为5 mL/kg。治疗试验先腹腔注射建立模型,后进行灌胃。检测小鼠血清AST、ALT酶含量、肝脏中抗氧化酶SOD、GSH-PX、脂质过氧化产物MDA;HE染色观察小鼠肝脏组织病理学变化。结果在预防实验中,与模型组相比,3组补硒的ALT、AST、MDA降低明显;低硒组SOD活性、中及高硒组GSH-PX活性显著性增加(P<0.05)。治疗试验中,与模型组相比,3组补硒组的ALT、AST活性未见明显差异,低、中硒组的MDA显著性降低,SOD显著性增加(P<0.05),中、高硒组GSH-PX活性显著性增加(P<0.05)。结论富硒小球藻提取物对急性肝损伤有一定的治疗和预防作用。 Objective To study the preventive and therapeutic effects of selenium-enriched chlorella extract(3069 mg/kg selenium content)on acute liver injury in mice.Methods 42 male C57 mice were selected for therapeutic and prevention experiments,and randomly divided into control group,model group,biphenyl diester drug group,selenium free group,low selenium group,medium selenium group,and high selenium group,with 10 rats in each group.In the prevention experiment,the control group and model group were given 20 mL/kg of distilled water by gavage daily,the drug group was given 0.200 g/kg of Bifendate Pills,the selenium supplement group was given 0.016,0.003,0.065 g/kg of selenium-enriched Chlorella,and the selenium free group was given 0.065 g/kg of Selenium-free Chlorella.Once a day,for 7 consecutive days,after the last gavage for 1 h,the control group was intraperitoneally injected with soybean oil,and the other groups were intraperitoneally injected with 10%CCl 4 soybean oil solution to establish a liver injury model,and the injection dose was 5 mL/kg.In the adjuvant therapy experiment,the model was established by intraperitoneal injection,and then administered by gavage.The number of animals,grouping,gavage dose and model establishment operation were the same as those of the prevention experiment mice.The contents of serum AST and ALT enzymes,antioxidant enzymes SOD,GSH-PX and lipid peroxidation product MDA in the liver were detected.Histopathological changes in the liver of mice were observed by HE staining.Results In the prevention experiment,compared with the model group,the ALT,AST and MDA of selenium in the three groups are significantly decreased.The SOD activity of the low-dose group and the GSH-PX activity of the middle and high-dose groups are significantly increased(P<0.05).In the treatment experiment,compared with the model group,there is no significant difference in ALT and AST activities in the three selenium supplementation groups,the MDA significance decreased in the low and medium selenium groups,the SOD significance increased(P<0.05),and the GSH-PX activity in the medium and high selenium groups increased significantly(P<0.05).Conclusion It shows that the selenium-enriched chlorella extract has certain therapeutic and prevention effects on acute liver injury.
