绞股蓝对环磷酰胺致免疫力低下小鼠的免疫调节及菌群紊乱调节 被引量:4

The effects of Gynostemma on cyclophosphamide-induced immunosuppression and dysbiosis in mice

摘要 目的探讨绞股蓝水提物对环磷酰胺诱导免疫力低下小鼠免疫力及菌群紊乱的影响。方法利用免疫抑制剂环磷酰胺诱导建立免疫低下小鼠模型。将21只SPF小鼠随机分为对照组、模型组和绞股蓝组,每组7只。建模后测定脾脏指数、脾脏淋巴细胞亚群水平,采用高通量测序技术分析各组小鼠结肠内容物菌群。结果绞股蓝水提液能增强免疫功能低下小鼠的免疫功能,显著提高免疫低下小鼠的脾脏指数及CD4^(+)T淋巴细胞水平,从而发挥免疫调节作用。绞股蓝水提液还可以显著降低小鼠肠道菌群中致病菌葡萄球菌的相对丰度,增加乳杆菌、拟杆菌等有益菌的相对丰度,从而发挥其免疫调节能力。结论绞股蓝水提液能够提高环磷酰胺诱导免疫力低下小鼠的免疫力,其机制可能与调节肠道微生态平衡密切相关。 Objective To explore the effect of water extract of Gynostemma on immunity and flora disorder in mice with cyclophosphamide induced immune deficiency.Methods Immunosuppressant cyclophosphamide was used to induce the immunosuppressive mouse model.Twenty-one SPF mice were randomly divided into the control group,model group and Gynostemma group(n=7).After modeling,the spleen index and lymphocyte subpopulations of the spleen were determined,and the colonic contents of the mice in each group were analyzed with high-throughput sequencing technology.Results The aqueous extract of Gynostemma enhanced the immune function of the immunocompromised mice,significantly increased the spleen index and CD4^(+)T lymphocyte content of the immunocompromised mice,and thus played an immunomodulatory role.The aqueous extract of Gynostemma also significantly reduced the relative abundance of pathogenic bacterium Staphylococcus in the intestinal flora of mice,and increased the relative abundance of beneficial bacteria such as Lactobacillus and Bacteroides,demonstrating an immune regulatory ability.Conclusion The aqueous extract of Gynostemma can improve the immunity of mice with cyclophosphamide induced immune deficiency,and the mechanism may be closely related to the regulation of intestinal microecological balance.
