摘要
目的通过分析miR-20b-5p、TGFBR2在宫颈癌组织中的表达水平及与临床病理特征的关系,探讨miR-20b-5p与TGFBR2在宫颈癌转移中的作用。方法选取2018年1月至2020年10月诊治的58例宫颈癌患者为宫颈癌组,选取同期32例因子宫肌瘤或子宫腺肌症行全子宫切除术患者的正常宫颈组织为对照组。RT-PCR检测2组宫颈组织中miR-20b-5p、TGFBR2 mRNA表达情况,免疫组化及Western blot检测宫颈癌组织中TGFBR2蛋白表达情况,通过在线网站预测miR-20b-5p与TGFBR2的靶向结合位点,并分析两者相关性,讨论两者其在宫颈癌组中表达差异与临床病理特征的关系。结果宫颈癌组中miR-20b-5p表达量显著高于对照组(P<0.01),而宫颈癌组中TGFBR2 mRNA表达量显著低于对照组(P<0.01)。免疫组化及Western blot结果进一步证实TGFBR2在宫颈癌组中表达水平显著低于对照组(P<0.01)。miR-20b-5p表达水平与TGFBR2 mRNA表达水平呈负相关(r=-0.7366,P<0.01)。且miR-20b-5p表达水平与宫颈癌患者的FIGO分期、淋巴结转移相关(P<0.05),与年龄、病理类型、分化程度、肿瘤大小无相关性(P>0.05);TGFBR2表达与宫颈癌患者的分化程度、FIGO分期、淋巴结转移相关(P<0.05),与年龄、病理类型、肿瘤大小无相关性(P>0.05)。结论miR-20b-5p与TGFBR2的异常表达可能参与了宫颈癌的发生发展,且两者存在密切联系,可能作为宫颈癌淋巴转移和预后的标志物。
Objective To analyze the expression levels of microRNA-20b-5p(miR-20b-5p)and Transforming growth factor beta receptorⅡ(TGFBR2)in cervical cancer tissues and their correlation with clinicopathological features,aiming to explore the role of miR-20b-5p and TGFBR2 in cervical cancer metastasis.Methods A total of 58 patients with cervical cancer who were diagnosed and treated from January 2018 to October 2020 were selected as the cervical cancer group,and 32 patients with normal hysterectomy due to uterine fibroids or adenomyosis during the same period were selected as the control group.Reverse-transcription polymerase chain reaction(RT-PCR)was performed to detect the mRNA expressions of miR-20b-5p and TGFBR2 in cervical tissues of both groups.The protein level of TGFBR2 in cervical cancer tissues was detected by immunohistochemistry and Western blot.The targeted binding site between miR-20b-5p and TGFBR2 was predicted by online website,and the correlation between the two was analyzed.The expression differences of miR-20b-5p and TGFBR2 in cervical tissues,and their correlation with clinicopathological characteristics of cervical cancer was discussed.Results MiR-20b-5p was significantly upregulated in the cervical cancer group than that in the control group,while mRNA expression of TGFBR2 was downregulated(all P<0.01).The immunohistochemistry and Western blot showed that the TGFBR2 was significantly downregulated in the cervical cancer group than that in the control group(P<0.01).The mRNA level of miR-20b-5p was negatively correlated with that of TGFBR2(r=-0.7366,P<0.01).The expression level of miR-20b-5p was significantly correlated with the Federation Internationale de Gynecologie et d’Obstetrique(FIGO)staging and lymph node metastasis of cervical cancer(P<0.05),which was not correlated with the age,pathological type,differentiation level and tumor size(P>0.05).The expression level of TGFBR2 was significantly correlated with the differentiation level,FIGO staging and lymph node metastasis of cervical cancer(P<0.05),which was not correlated with the age,pathological type and tumor size(P>0.05).Conclusion Abnormally expressed miR-20b-5p and TGFBR2 may be involved in the occurrence and development of cervical cancer,showing a close correlation.They may serve as biomarkers for lymph node metastasis and prognosis of cervical cancer.
