摘要
通过研究岩藻黄素(Fucoxanthin,FUCO)对H_(2)O_(2)诱导斑马鱼(Barchydanio rerio)衰老的抑制作用,为FUCO药用价值研究和抗衰老药物研发提供理论基础。本研究采用H_(2)O_(2)诱导建立斑马鱼衰老模型,用岩藻黄素进行干预,测定斑马鱼胚胎存活率、体内SAβ半乳糖苷酶活性、丙二醛(Malondialdehyde,MDA)含量、活性氧(Reactive Oxygen Species,ROS)含量、超氧化物歧化酶(Superoxide Dismutase,SOD)活性,探讨FUCO的抗衰老作用。结果显示,2 mmol/L的H_(2)O_(2)作用于斑马鱼胚胎96 h可诱导斑马鱼胚胎衰老;与衰老模型组(M)相比,4个FUCO处理组(2.5、5.0、10.0、15.0μmol/L)斑马鱼胚胎体内的SAβ半乳糖苷酶活性均降低,MDA含量和ROS含量均明显下降,SOD活性均增高。本研究表明,浓度为2.5-15.0μmol/L时,FUCO对H_(2)O_(2)诱导斑马鱼衰老具有较好的抑制作用。
By studying the inhibitory effect of fucoxanthin(FUCO)on H_(2)O_(2)induced senescence in Barchydanio rerio,it provided a theoretical basis for the study of fucoxanthin medicinal value and the development of anti aging drugs.In this study,the aging model of H_(2)O_(2)induced Barchydanio rerio was established,and the fucoxanthin was used to intervene.The survival rate,SA-β-galactosidase activity,malondialdehyde(MDA)content,reactive oxygen species(ROS)content,and superoxide dismutase(SOD)activity of Barchydanio rerio were measured to investigate the anti aging effect of FUCO.The results showed that the senescence of Barchydanio rerio could be induced by 2 mmol/L H_(2)O_(2)treatment for 96 h.Compared with the aging model group(M),the SA-β-galactosidase activity in the four FUCO treatment groups(2.5μmol/L,5.0μmol/L,100μmol/L,15.0μmol/L)decreased,and the contents of MDA and ROS significantly decreased,while the SOD activity increased.The result of this study shows that FUCO has a good inhibitory effect on H_(2)O_(2)induced senescence in Barchydanio rerio,when the concentration is 2.5-15.0μmol/L.
