摘要
目的:探讨TRIM32在肝细胞癌(HCC)组织中的表达和预后价值,及其对HCC细胞恶性生物学行为的影响。方法:使用癌症基因组图谱(TCGA)的测序数据评估HCC中TRIM32的表达是否有差异。qPCR和Western Blot分别检测TRIM32在HCC组织中的mRNA和蛋白表达水平。进一步分析TRIM32的表达水平与HCC患者临床病理特征的关系。使用Kaplan-Meier Plotter网站和Cox回归分析评估TRIM32在HCC中的预后价值。向HCC细胞中转染si-TRIM32,分别采用细胞增殖实验、划痕实验和Transwell侵袭实验检测敲低TRIM32对肝癌细胞增殖、迁移和侵袭的影响。利用Linkedomics数据库分析与TRIM32相关的基因并进行KEGG分析。结果:TRIM32在HCC组织高表达(P<0.05)。TRIM32表达的上调与HCC患者的病理分期、组织学分级、血清甲胎蛋白(AFP)表达升高和不良预后显著相关(P<0.05)。功能实验表明,敲低TRIM32可以抑制HCC细胞的增殖、迁移和侵袭(P<0.05)。KEGG分析表明,TRIM32及其相关基因在甲状腺激素信号通路、TGF-β信号通路和Hippo信号通路中显著富集。结论:TRIM32在HCC组织中高表达且与患者不良预后显著相关,敲低TRIM32表达能够抑制HCC细胞的恶性生物学行为。
Objective:To explore the expression and prognostic value of TRIM32(tripartite motif containing 32)in hepatocellular carcinoma(HCC)tissues,and its influence on the malignant biological behaviors of HCC cells.Methods:Sequencing data from The Cancer Genome Atlas(TCGA)were used to assess whether there were differences in TRIM32 expression in HCC.The mRNA and protein expression levels of TRIM32 in HCC tissues were detected by qPCR and Western Blot assay,respectively.The relationship between the expression level of TRIM32 and the clinicopathological characteristics of HCC patients was further analyzed.Kaplan-Meier Plotter website and Cox regression analy-sis were used to evaluate the prognostic value of TRIM32 in HCC.After transfection of si-TRIM32 into HCC cells,the effects of TRIM32 on the proliferation,migration,and invasion of HCC cells were detected by cell proliferation assay,scratch assay,and Transwell invasion assay,respectively.The genes related to TRIM32 were analyzed using the Linkedomics database,and KEGG analysis was performed.Results:TRIM32 was overexpressed in HCC tissues(P<0.05 or P<0.01).The upregulation of TRIM32 was significantly associated with pathologic staging,histological grade,elevated serum alpha-fetoprotein(AFP)expression,and poor prognosis in HCC patients(P<0.05).Functional experiments showed that knockdown of TRIM32 could inhibit the proliferation,migration,and invasion of HCC cells(P<0.05).KEGG analysis showed that TRIM32 and its related genes were significantly enriched in thyroid hormone signaling pathway,TGF-βsignaling pathway,and Hippo signaling pathway.Conclusion:TRIM32 is highly expressed in HCC tissues and cells,and is significantly related to the poor prognosis of patients;knockdown of TRIM32 can inhibit the malignant biological behaviors of HCC cells.
