摘要
目的:泛癌症分析肌动蛋白结合蛋白(ANLN)基因的表达、预后及其潜在作用机制。方法:应用TCGA及GTEx数据库及Oncomine和TIMER网站泛癌症分析ANLN的表达变化及其与预后的关系;通过Coexpedia网站获取ANLN的共表达基因,应用Metascape网站进行基因本体(GO)和KEGG信号通路分析;通过R软件分析获得ANLN与DNA甲基化的相关性。结果:ANLN在肾上腺皮质癌、乳腺癌及结直肠癌等26种肿瘤中高表达(P<0.001),而在急性髓细胞样白血病中低表达(P<0.001)。ANLN表达不利于肾上腺皮质癌、膀胱癌、宫颈鳞癌和腺癌等13种肿瘤的预后(P<0.05),而利于胸腺癌的预后(P=0.00053)。ANLN的共表达基因主要富集于细胞分裂、DNA复制及细胞周期相变等基因模块;其参与的信号通路主要包括细胞周期、DNA复制及p53信号通路等。此外,泛癌症分析显示ANLN与DNA甲基化过程中的甲基转移酶具有一定相关性。结论:ANLN在泛癌分析中普遍高表达,而在急性髓细胞样白血病中低表达。且ANLN表达利于胸腺癌的预后,而不利于肾上腺皮质癌、膀胱癌、宫颈鳞癌和腺癌等13种肿瘤的预后,可作为上述肿瘤的潜在预后标志物及治疗靶点。
Objective:To analyze the expression,prognosis,and potential mechanism of actin binding protein(ANLN)gene in pan-cancer.Methods:The expression and prognosis of ANLN were analyzed by TCGA and GTEx databases,and through Oncomine and TIMER websites.The co-expressed genes of ANLN were obtained from Coexpedia website,and the gene ontology(GO)and KEGG signal pathway were analyzed by Metascape website.The correlation between ANLN and DNA methylation was obtained by R software analysis.Results:ANLN was highly expressed in 26 tumors,including adrenal cortical carcinoma,breast cancer,and colorectal cancer(P<0.001),and poorly expressed in acute myeloid leukemia(P<0.001).The expression of ANLN was detrimental to the prognosis of 13 tumors,including adrenal cortical carcinoma,bladder carcinoma,cervical squamous cell carcinoma,and adenocarcinoma(P<0.05),and beneficial to the prognosis of thymic carcinoma(P=0.00053).The co-expressed genes of ANLN were mainly concentrated in cell division,DNA replication,and cell cycle phase transition.The signaling pathways involved mainly included cell cycle,DNA replication and p53 signaling pathway.In addition,pan-cancer analysis showed a correlation between ANLN and methyltransferase in DNA methylation.Conclusion:The expression of ANLN was high in panocarcinoma and low in acute myeloid leukemia.Moreover,the expression of ANLN is favorable for the prognosis of thymic carcinoma,but unfavorable for the prognosis of adrenal cortical carcinoma,bladder carcinoma,cervical squamous cell carcinoma,and adenocarcinoma,etc.,so it can be used as a potential prognostic marker and therapeutic target for these tumors.
