摘要
目的 探究基于中药金汁苦寒药性粪菌移植(FMT)干预湿热型溃疡性结肠炎(UC)的作用机制。方法 40只小鼠随机分为正常对照组、湿热型模型组、湿热型FMT组和湿热型5-氨基水杨酸(5-ASA)组,各10只,比较各组小鼠体质量、结肠长度、结肠质量、疾病活动指数(DAI)评分及结肠黏膜病理形态学差异,并检测各组小鼠结肠组织中相关细胞因子水平及m RNA表达情况。结果 湿热型FMT组、湿热型5-ASA组体质量、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)和转化生长因子β(TGF-β)水平以及IL-4、IL-10和TGF-β m RNA表达高于湿热型模型组(P <0.05),结肠长度长于湿热型模型组(P <0.05),结肠质量、结肠质量指数、DAI评分、病理评分、干扰素-γ(INF-γ)、白细胞介素-2(IL-2)和白细胞介素-17(IL-17)水平以及INF-γ、IL-2和IL-17 m RNA表达低于湿热型模型组(P <0.05)。结论 中药金汁FMT对湿热型UC有一定治疗作用,可能是通过调节免疫系统平衡来发挥作用。
Objective To explore the role mechanism of fecal microbiota transplantation(FMT)on the intervention of ulcerative colitis(UC)of dampness-heat syndrome based on the bitter-cold medicinal properties of Chinese medicine Jinzhi.Methods 40 mice were randomly divided into normal control group,damp-heat model group,damp-heat FMT group and damp-heat 5-ASA group,with 10 mice in each group.Body mass,colon length,colon mass,DAI score and pathological morphology of colon mucosa were compared among the groups.The levels and mRNA expressions of related cytokines in the colon tissues of each group of mice were detected.Results The body mass,levels of IL-4,IL-10 and TGF-βand mRNA expressions of IL-4,IL-10 and TGF-βin damp-heat FMT group and damp-heat 5-ASA group were heavier or higher than those in damp-heat model group(P<0.05),the colon length was longer than that in damp-heat model group(P<0.05),colon mass,colon mass index,DAI score and pathology score,levels of INF-γ,IL-2 and IL-17 and mRNA expressions of INF-γ,IL-2 and IL-17 were lighter or lower than those in damp-heat model group(P<0.05).Conclusion FMT based on traditional Chinese medicine Jinzhi has a certain therapeutic effect on UC of damp-heat syndrome,and may improve the UC of damp-heat syndrome by regulating the balance of immune system.
