金匮肾气丸和桂附地黄丸治疗慢性肾小球肾炎作用机制探讨

Mechanism of Jingui Shenqi Pill and Guifu Dihuang Pill in Treating Chronic Glomerulonephritis

目的:基于网络药理学探讨金匮肾气丸和桂附地黄丸二者治疗慢性肾小球肾炎(Chronic glomerulonephritis,CGN)的作用靶点以及作用机制。方法:利用TCMSP数据库筛选金匮肾气丸和桂附地黄丸的活性成分和中药靶标,使用Uniprot数据库注释基因名,并在GeneCards等数据库中检索CGN的疾病靶标。利用Cytoscape软件,构建药物-有效成分-靶点-疾病网络图,将数据导入String数据库,从而探究蛋白之间的作用关系。最后利用R语言进行基因本体功能注释及KEGG通路富集分析。结果:分别筛选得到金匮肾气丸和桂附地黄丸核心活性成分139和95个,治疗CGN的靶标分别为111和58个;二者均涉及AGE-RAGE、脂质和动脉粥样硬化、乙肝、化学致癌-受体激活、PI3K/Akt、TNF等信号通路。结论:运用网络药理学探究金匮肾气丸和桂附地黄丸对CGN的治疗机制,发现二者大部分相同,但金匮肾气丸作用靶点更多,且调控的信号通路更多在下游,为进一步试验验证提供理论基础。

Objective:Based on network pharmacology,to explore the targets and mechanisms of Jinkui Shenqi pills and Guifudihuang pills in the treatment of chronic glomerulonephritis(CGN).Methods:The TCMSP database was used to screen the active ingredients and TCM targets of Jinkui Shenqi Pills and Guifu Dihuang Pills,the gene names were annotated using the Uniprot database,and the disease targets of CGN were searched in databases such as GeneCards.Use Cytoscape software to build a drug-active ingredient-target-disease network diagram,and import the data into the String database to explore the relationship between proteins.Finally,R language was used for gene ontology function annotation and KEGG pathway enrichment analysis.Results:139 and 95 core active ingredients of Jinkui Shenqi Pills and Guifu Dihuang Pills were screened,respectively,and 111 and 58 targets for treating CGN were obtained;both involved AGE-RAGE,lipids and atherosclerosis,hepatitis B,chemical carcinogenesis-receptor activation,PI3K/Akt,TNF and other signaling pathways.Conclusion:Using network pharmacology to explore the therapeutic mechanism of Jingui Shenqi Pills and Guifu Dihuang Pills on CGN,it is found that the two are mostly the same,but Jinkui Shenqi Pills have more targets,and more signaling pathways are downstream,to provide a theoretical basis for further experimental verification.
