干鱼腥草及鲜鱼腥草混合绿茶颗粒抗糖尿病作用及机制初探

Preliminary Study on Antidiabetic Effect and Mechanisms of Dried and Fresh Houttuynia cordata Mixed with Green Tea Granules

摘要 目的:研究干鱼腥草、鲜鱼腥草混合绿茶粉末的代用茶颗粒制剂对糖尿病模型小鼠的作用,及两种制剂对α-淀粉酶、α-葡萄糖苷酶的体外抑制作用。方法:将实验小鼠随机分为正常组、模型组、干鱼腥草+绿茶代用茶颗粒组及鲜鱼腥草+绿茶代用茶颗粒组;除正常组外的小鼠采用四氧嘧啶法建立糖尿病小鼠模型。将含干鱼腥草、鲜鱼腥草的代用茶颗粒剂量折算后进行日常小鼠灌胃并饲养一段时间后,测定空腹血糖值、血清胰岛素浓度。将含干鱼腥草及鲜鱼腥草颗粒配制成不同浓度,采用碘比色法、ELISA法测定其体外对α-淀粉酶、α-葡萄糖苷酶的抑制率。结果:糖尿病小鼠模型与正常小鼠相比,空腹血糖值升高、血清胰岛素浓度下降(P<0.05);两种鱼腥草+绿茶组能明显降低糖尿病模型小鼠的空腹血糖值,且升高血清胰岛素浓度(P<0.05)。含干鱼腥草、鲜鱼腥草的代用茶颗粒均无明显的α-淀粉酶抑制作用,有明显的α-葡萄糖苷酶抑制作用;含鲜鱼腥草颗粒在高浓度时对α-葡萄糖苷酶抑制率高于干鱼腥草(P<0.05)。结论:干、鲜鱼腥草+绿茶的代用茶颗粒能有效控制多项与糖尿病相关的指标,可以作为糖尿病控制及辅助治疗的选择之一,具有良好的应用前景。 Objective:To study the effects of dried fishy grass and fresh fishy grass mixed with green tea powder in the preparation of tea granules on diabetic model mice,and the inhibition ofα-amylase andα-glucosidase by the two preparations in vitro.Method:Experimental mice were randomly divided into groups:normal,model,dried Houttuynia cordata and fresh Houttuynia cordata.Mice were prepared as diabetic model mice by alloxan except mice in normal group.The dosage of dried and fresh Houttuynia cordata granule herb tea was converted into mice dosage.After feeding for a period,measured fasting blood sugar and serum insulin concentration of mice in different groups.The granules containing dried and fresh Houttuynia cordata were prepared into different concentrations;the inhibition rates ofα-amylase andα-glucosidase in vitro experiment on were tested by iodine colorimetry and ELISA methods.Result:After feeding for several days,compared with normal mice,the fasting blood glucose of diabetic model mice increased and the serum insulin concentration decreased(P<0.05);granule herb tea containing dried and fresh Houttuynia cordata could significantly reduce the fasting blood glucose of diabetic mice,and increase the serum insulin concentration(P<0.05).Dried and fresh Houttuynia cordata granule herb tea have no obvious inhibition effect ofα-amylase and obvious inhibition effect ofα-glucosidase.The inhibition rate of fresh Houttuynia cordata onα-glucosidase at high concentration is higher than that of corresponding dried group.Conclusion:The tea substitute granules of dried and fresh fishy grass+green tea can effectively control several indicators related to diabetes and can be one of the options for diabetes control and adjuvant therapy,which has good application prospects.
