摘要
建立高效液相色谱同时测定复方南板蓝根片中秦皮乙素和菊苣酸含量的方法。采用XBridge C18色谱柱(250 mm×4.6 mm,5μm)分离,流动相为乙腈-0.3%磷酸溶液,梯度洗脱,流量为1 mL/min,检测波长为330 nm,柱温为40℃,进样体积为10μL,以色谱峰面积外标法定量。秦皮乙素和菊苣酸的质量浓度分别在18.40~184.02、4.96~49.61 g/mL范围内与色谱峰面积的线性关系良好,相关系数均为0.9999。秦皮乙素、菊苣酸测定结果的相对标准偏差分别为0.72%、0.54%(n=6),平均回收率分别为97.40%、98.45%。该方法操作简单,重复性好,准确度高。
A high performance liquid chromatography(HPLC)method was established for simultaneous determination of aesculetin and cichoric acid in Compound Banlangen Tablets.XBridge C18 chromatographic column(250 mm×4.6 mm,5μm)was used,the mobile phase was acetonitrile-0.3%phosphoric acid solution with gradient elution and the flow rate of 1 mL/min.The detection wavelength was 330 nm,the column temperature was 40℃,the injection volume was 10μL,and external standard method was used for quantitative.The linear ranges of aesculetin and cichoric acid with the chromatographic peak area were 18.40-184.02µg/mL(r=0.9999)and 4.96-49.61µg/mL(r=0.9999),respectively.The relative standard deviations of aesculetin and cichoric acid determination results were 0.72%and 0.54%(n=6),respectively,the average recoveries were 97.40%and 98.45%,respectively.The method is simple,reproducible and accurate.
