Quality Evaluation of Atractylodis Macrocephalae Rhizome from Different Origins Based on Quantitative Analysis of Multi-components by Single Marker

摘要 目的建立一测多评(QAMS)法同时测定白术药材中白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ和苍术酮的含量,并对挥发油和醇溶性浸出物进行测定,评价不同产地白术药材的质量。方法采用Waters Sunfire C_(18)色谱柱,流动相为乙腈和水,梯度洗脱,流速为1.0 mL/min,柱温为25℃,检测波长为220 nm和276 nm。以白术内酯Ⅲ为内参物,测定白术内酯Ⅱ、白术内酯Ⅰ和苍术酮的相对校正因子,计算各成分含量,同时与外标法(ESM)实测值进行比较,以验证QAMS法的可行性;参照2020年版《中国药典》对不同产地白术药材的挥发油、醇溶性浸出物含量进行测定。结果25批不同产地白术药材中各成分QAMS计算值与ESM实测值无显著差异。浙江产地的白术中苍术酮和挥发油含量高于其它产地。结论所建立的HPLC-QAMS法为白术药材提供了一种准确、低成本的多指标质量评价方法。挥发油可作为浙产白术的道地质量评价指标。 Objective To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous determination of atractylenolideⅢ,atractylenolideⅡ,atractylenolideⅠand atractone in Atractylodis macrocephalae rhizome,and the determination of essential oil and alcohol-soluble extractives to evaluate the quality of Atractylodis macrocephalae rhizome from different origins.Methods The chromatography was performed on a Waters Sunfire C_(18) column with the mobile phases of acetonitrile and water in gradient elution at a flow rate of 1.0 mL/min and a column temperature of 25℃with the detection wavelengths of 220 nm and 276 nm.The relative calibration factors of atractylenolideⅢ,atractylenolideⅡ,atractylenolideⅠand atractylone were determined and the contents of each component were calculated by using atractylenolideⅢas the internal reference,while the measured values were compared with the external standard method(ESM)to verify the feasibility of the QAMS method.The contents of essential oil and alcohol-soluble extractives of Atractylodis macrocephalae rhizome from different origins were determined with reference to the 2020 edition of the Chinese Pharmacopoeia.Results There was no significant difference between the calculated values of each component in 25 batches of Atractylodis macrocephalae rhizome from different origins by QAMS and the actual measured values by ESM.Atractone and essential oil contents in Atractylodis macrocephalae rhizome of Zhejiang origin were higher than those of other origins.Conclusion The established HPLC-QAMS method provides an accurate and low-cost multi-indicator quality evaluation method for Atractylodis macrocephalae rhizome.The essential oil can be used as potential authentic markers of Zhejiang Atractylodis macrocephalae rhizome.
