彩色花椰菜叶绿体全基因组特征分析 被引量:1

Characterization of the Complete Chloroplast Genome of Brassica oleracea L.var.botrytis

摘要 使用Illumina HiSeq 2500高通量测序平台测序组装白色、紫色、橙色、淡绿色4种花椰菜叶绿体全基因组,分析结果显示,花椰菜叶绿体基因组全长为153 364 bp(淡绿色)和153 366 bp(白色、紫色、橙色),由4个区域组成,包括1个88 136~88 138 bp的长单拷贝区(LSC)、1个17 834~17 835 b的短单拷贝区(SSC)以及2个26 196~26 197 bp的反向重复(IRa/IRb),4种花椰菜叶绿体基因组均注释132个基因,包括87个蛋白编码基因、37个tRNA和8个rRNA。有19个基因在IR区重复;平均总GC含量均为36.36%;共有67种密码子编码24种氨基酸,32个密码子的使用频率(RSCU)大于1.00,偏向于以A或U结尾的同义密码子占90.6%,系统发育分析表明,花椰菜和甘蓝亲缘关系最近,它们与芥菜和欧洲油菜聚为一大类。 The complete chloroplast genome of 4 varieties of Brassica oleracea L.var.botrytis including white,purple,orange and light green types were sequenced and assembled by Illumina HiSeq 2500 sequencing platform,the analysis results showed that the whole chloroplast genome of Brassica oleracea L.var.botrytis were 153364 bp(light green)and 153366 bp(white,purple,orange),formed by four regions,including a large single copy region(LSC)with 88136-88138 bp,a small single copy region(SSC)with 17834-17835 bp,two inverted repeat regions(IRa/IRb)with 26196-26197 bp.The chloroplast genome of four varieties of Brassica oleracea L.var.botrytis encoded 132 genes,including 87 protein-coding genes,37 tRNA genes,and 8 rRNA genes,with 19 duplicates in IRs.The total GC content of the entire chloroplast genome accounts for 36.36%.A total of 67 codons encoded 24 kinds of amino acids.The RSCU values for 32 codons were over 1.00,and 90.6%of the codons ended with A or U.The phylogenetic analysis suggested that Brassica oleracea L.var.botrytis was closely related to Brassica oleracea L.var.capitate and Brassica oleracea L.var.italic.These were grouped together with Brassica juncea and Brassica napus.
