摘要
氯硝柳胺用于绦虫病治疗已有50多年历史,最新研究表明其对白血病等肿瘤、炎症相关疾病和新型冠状病毒肺炎等均具有一定疗效,具有很高的老药新用价值。本综述总结了氯硝柳胺在抗肿瘤、炎症相关疾病和病毒感染3类疾病中的研究进展,并从NF-κB、Wnt/β连环蛋白、Notch、信号转导和转录激活因子3和哺乳动物西罗莫司(雷帕霉素)靶蛋白复合物1等5种信号通路分析其多种药理作用及其诱导肿瘤细胞凋亡、降低炎症及抑制肿瘤和病毒扩增的分子机制。
Niclosamide has been widely used in the treatment of tapeworm for more than 50 years.The latest studies show that it is an old drug that can be novelly used in treating tumors,inflammatory disease,and even viral infections.The present review summarizes the current progress in research on the efficacy of niclosamide in such diseases as tumors,inflammatory diseases and viral infections.It also discusses the diverse pharmacological activities and anti-tumor/inflammation/virus molecular mechanisms of niclosamide from the perspectives of nuclear factor-kappa B,Wnt/β-catenin,Notch,signal transducers and activators of transcription 3 and mammalian targets of rapamycin complex 1.
