胺功能化SBA-15固定碳酸酐酶的制备及其性能研究

摘要 利用水热法制备介孔球形SBA-15并进行胺功能化,以其为载体通过交联酶法将碳酸酐酶固定在载体上,得到固定化碳酸酐酶。通过比表面积(BET)、扫描电镜(SEM)、X射线衍射(XRD)、傅里叶红外(FTIR)、核磁共振(NMR)等手段对载体材料和固定化酶的物理化学性质进行表征分析。采用量热法测定碳酸酐酶活力,并通过酶活回收率评价碳酸酐酶的固定效果。结果表明,用该方法能制备介孔球形SBA-15,并能成功对其进行胺功能化得到APTES/SBA-15,以SBA-15和APTES/SBA-15为载体得到的固定化碳酸酐酶的酶活回收率分别达到36%和52%。 Mesoporous spherical SBA-15 was prepared by hydrothermal method and functionalized with amines.Carbonic anhydrase was immobilized on the carrier by cross-linking enzyme method.The carrier materials and immobilized enzymes were characterized by surface area(BET),scanning electron microscopy(SEM),X-ray diffraction(XRD),fourier transform infrared(FTIR)and nuclear magnetic resonance(NMR),and their physical and chemical properties were compared.The activity of carbonic anhydrase was measured by calorimetry,and the fixation effect of carbonic anhydrase was evaluated by activity recovery.The results show that mesoporous spherical SBA-15 can be prepared by this method,and APTES/SBA-15 can be successfully functionalized with amines.The enzyme activity recovery of immobilized carbonic anhydrase using SBA-15 and APTES/SBA-15 as carriers is 36%and 52%,respectively.
