西兰花毛状根次生代谢物与信号分子对外源褪黑素的生理响应

Physiological Response of Secondary Metabolites and Signaling Molecules to Melatonin in Broccoli Hairy Roots

摘要 为了探究外源褪黑素(melatonin,MT)对西兰花毛状根中萝卜硫苷(glucoraphanin,GRA)、萝卜硫素(sulforaphane,SF)、信号分子NO和H_(2)O_(2)含量及黑芥子酶(myrosinase enzyme,MYR)活力的影响,以发根农杆菌(Agrobacterium rhizogenes)‘ATCC15834’侵染西兰花‘中青9号’获得的毛状根为实验材料,在探究西兰花毛状根中次生代谢物GRA和SF合成动态的基础上添加不同浓度的MT处理,检测GRA及SF产量,NO和H_(2)O_(2)含量与MYR活力。结果表明,未添加外源MT时,西兰花毛状根生长至20 d GRA和SF产量达到最大;经500μmol/L MT处理后的西兰花毛状根培养体系GRA和SF总产量均达最高,分别为2 324.44和656.13μg/瓶,分别是对照的1.30和1.31倍,同时提高了MYR的活力(0.16 U/g),为对照的1.16倍,不同浓度MT处理下,MT处理浓度为500μmol/L时对毛状根中NO含量影响最为显著(P≤0.05),为0.40μmol/g,是对照组的2.23倍;MT处理浓度为1 000μmol/L时,H_(2)O_(2)含量最高,达4.62μmol/g。本研究结果为利用MT诱导植物生产次生代谢物的研究提供了基础数据。 In order to explore the effects of exogenous melatonin(MT)on the contents of glucoraphanin(GRA),sulforaphane(SF),NO and H,O and myrosinase enzyme(MYR),the hairy roots of broccoli'Zhongqing9'infected by A grobacterium rhizoge nes'ATCC15834'were used as experimental materials.On the basis of exploring the synthesis dynamics of secondary metabolites GRA and SF in hairy roots of broccoli,MT treatment with different concentra-tions was added to detect GRA and SF production,NO and H,O2 contents and MYR activity.The results showed that the yield of secondary metabolites GRA and SF reached the maximum on the 20 d of the growth of broccoli hairy roots.The total yield of GRA and SF in the culture system of broccoli hairy reached the maximum after MT treatment with 500μmol/L compared with the control,respectively,which were 2324.44 and 656.13μg/flask fold of that of the control which were 1.30 and 1.31 fold of the control.Besides,the activity of MYR was increased by 0.16 U/g,which was 1.16 fold of the control.Under different MT treatment,NO synthesis were significantly promoted by 0.40μmol/g after MT(500μmol/L)treatments in hairy roots of broccoli(P≤0.05),which was 2.23 fold compared with the control group.When MT concentration was 1000μmol/L,the highest content of H,O2 was 4.62μmol/L.The results of this study provide basic data for the study of secondary metabolite production in plants induced by MT.
